摘  要:目的:探讨半枝莲水提液(SEB)和去纳米颗粒水提液(SEB-RN)体内外的抗肝肿瘤作用。方法:采用体外抑瘤实验培养人肝癌细胞SMMC-772,通过噻唑蓝(MTT)法检测其体外肿瘤抑制率;体内实验采用肝癌H22移植瘤法,以给药前后体质量、肿瘤抑制率、脾脏指数和胸腺指数为指标,对比SEB和SEB-RN的肿瘤抑制作用。结果:体外试验结果表明,环磷酰胺(CTX)组、SEB组和SEB-RN组均对SMMC-7721细胞的增殖有显著抑制作用;与SEB-RN组比较,SEB组对SMMC-7721细胞的增殖抑制率明显更高。体内试验结果表明,与模型组比较,CTX组、SEB组和SEB-RN组均能显著抑制实体瘤生长,与SEB-RN组比较,SEB组肿瘤抑制率明显更高;与模型组比较,CTX组胸腺、脾脏指数明显更低,SEB组、SEB-RN组明显更高,差异均有极显著统计学意义(P<0.01);SEB组胸腺、脾脏指数明显高于SEB-RN组,差异有统计学意义(P<0.05)。结论:SEB和SEB-RN均有一定的抗肿瘤和增强免疫功能的作用,但SEB优于SEB-RN。OBJECTIVE: To probe into the effects of water-extraction of scutellaria barbata( SEB) and waterextraction of scutellaria barbata with nanoparticles removed( SEB-RN) on Anti-liver tumor in vitro and in vivo.METHODS: In vitro tumor inhibition test was performed to culture SMMC-772,and MTT method was used to determine the proliferation of SMMC-772 in vitro. Liver cancer H22 transplantation tumor method was adopted for in vivo experiment; body mass,tumor inhibitory rate,spleen index and thymus index before and after administration were considered as indicators,the inhibitory effect on tumor was compared between SEB and SEB-RN. RESULTS: In vitro test show that: the CTX group,SEB group and SEB-RN group had significant inhibitory effects on the proliferation of SMMC-772; and the SEB group was higher than the SEB-RN group. In vivo test show that: compared with the model group,CTX group,SEB group and SEB-RN group could significantly inhibit the growth of solid tumor,and SEB group was significantly higher than the SEB-RN group. Compared with the model group,the thymus and spleen index of CTX were significantly lower,yet the data in SEB group and SEB-RN group were significantly higher,the difference was statistically significant( P〈0. 01); the thymus and spleen index of SEB group were significantly higher than those of SEB-RN group,with statistically significant difference( P〈0. 05). CONCLUSIONS: SEB and SEB-RN both have certain anti-tumor effects and improvement effects on immune function,yet the SEB is superior to SEB-RN.
