摘 要:为研究鸡线粒体抗病毒信号蛋白(mitochondrial anti-viral signaling protein,MAVS)在I型干扰素信号通路中的作用,对克隆的鸡MAVS基因进行了生物信息学分析。结果表明,多数在哺乳动物MAVS中发挥重要作用的蛋白基序及氨基酸位点在鸡中不保守;对鸡MAVS与其他物种进行了进化分析,发现该基因具有较强的种间特异性;DF-1细胞在感染新城疫病毒后12h,MAVS和IFN-β的mRNA水平均被上调,提示鸡MAVS可能参与了细胞抗病毒免疫反应;同时在DF-1细胞中过表达鸡MAVS能够激活IFN-β启动子活性。上述研究揭示了鸡MAVS在诱导I型干扰素产生中具有重要作用,为进一步研究禽类先天性免疫抗病毒途径奠定基础。In this study,the function of chicken mitochondrial anti-viral signaling protein(MAVS)in the type Ⅰ IFN signaling pathway was investigated.Alignment of the chicken MAVS(chMAVS)deduced amino acid sequence indicated that the major functional domains are not conserved in chMAVS compared with that of mammalian MAVS.Phylogenetic analysis demonstrated that chMAVS exhibited a strong evolutionary relationship with turkey and quail MAVS.Furthermore,levels of chMAVS and IFN-β mRNA increased in DF-1 cells infected with Newcastle disease virus(NDV),and reaching their highest level at 12h.Overexpression of chMAVS induced IFN-β activation in DF-1 cells.Together,these results indicate that chicken MAVS played a critical role in inducing IFN-β,and laid foundations for the further study of avian innate antiviral immune pathway.
