摘　　要：目的探讨程氏蠲痹汤加减方(CJBD)对佐剂性关节炎(AA)大鼠T淋巴细胞中的环腺苷酸(c AMP)水平和CD4^+/CD8^+T细胞比值的影响。方法将成年雄性SD大鼠分为正常对照组、AA组、CJBD组和雷公藤多苷片(TGT)组。用吹风冷水和Freund完全佐剂制备大鼠风寒湿痹AA模型,对照组给予生理盐水灌胃,其他组分别给予对应药品,持续7 d。各组大鼠经眼眶取血,分离外周血单个核细胞(PBMC),利用免疫磁珠分选技术分选T淋巴细胞,应用c AMP检测试剂盒检测各组T淋巴细胞的c AMP水平,用流式细胞术检测各组大鼠外周血T细胞中的CD4^+和CD8^+T细胞数,计算CD4^+/CD8^+T细胞比值。结果与正常对照组比较,CJBD组大鼠T淋巴细胞的c AMP表达水平显著降低,TGT组的c AMP含量与正常对照组无显著性差异;CJBD组的CD4^+/CD8^+T细胞比值显著降低。结论 CJBD可降低AA大鼠T淋巴细胞c AMP的水平和CD4^+/CD8^+T细胞比值。Objective To investigate the effects of modified Cheng＇s Juanbi Decoction （CJBD） on the level of cAMP in T lymphocytes and CD4^＋/CD8^＋ T cell ratio among rats with adjuvant arthritis （AA）. Methods Male adult SD rats were randomly divided into normal control group, AA group, CJBD group, and tripterygium glycosides tablet （TGT） group. Blowing/cold water and Freund＇s complete adjuvant were used to establish a rat model of AA with wind-cold-dampness arthralgia. The control group was given normal saline by gavage, and the CJBD group and TGT group were given respective therapies. The course of treatment was 7 days for all groups. Blood was collected from the orbit, and peripheral blood mononuclear cells （PBMCs） were isolated. Magnetic activated cell sorting was used to separate T lymphocytes. A cAMP detection kit was used to measure the cAMP levels in T lymphocytes. Flow cytometry was performed to determine the numbers of CD4^＋and CD8^＋ T cells in peripheral T cells, and the CD4^＋/CD8^＋ T cell ratio was calculated. Results Compared with the normal control group, the CJBD group had significantly reduced expression of cAMP in T lymphocytes, but there was no significant difference in cAMP level between the TGT group and the normal control group. The CJBD group had a significantly lower CD4^＋/CD8^＋ T cell ratio than the normal control group. Conclusion CJBD can reduce the cAMP level in T lymphocytes and CD4^＋/CD8^＋ T cell ratio in rats with AA.
