摘 要:目的研究不同时间点锰暴露对大鼠精子质量及血清性激素的影响。方法健康成年雄性SD大鼠96只,在严格控制光照/黑暗(12h:12h)条件下适应性喂养7天后,随机分为4个对照组与4个锰暴露组,分别于一天中的ZT2、ZT8、ZT14与ZT20授时时间点对锰暴露组腹腔注射30mg/kg MnCl_2,对照组等容量腹腔注射蒸馏水,每天1次,连续21天。第22天,处死动物,获取动物附睾、血清,检测精子质量、血清雌二醇(E2)、促卵泡激素(FSH)和睾酮(T)含量。结果锰暴露可引起精子活动度和精子数量减少,导致血清E2、FSH与T的紊乱,且具有时间差异性。其中,精子数量在各时间点暴露均下降,而精子活动度仅在ZT2和ZT8授时时间点暴露下降;E2升高,但仅表现在ZT2时间点暴露;FSH在ZT8、ZT14与ZT20授时时间点暴露下降;T在ZT2与ZT20时间点暴露降低,而在ZT14时间点暴露升高。结论锰暴露引起的精子损伤及激素分泌紊乱均具时间差异性,在锰的毒性评价时需考虑其时间差异性。Objective To explore the chronotoxicity effect of exposure to manganese (Mn) on the quantity of sperm and reproductive hormones in serum in male rats. Methods Ninety - six healthy male SD rats of SPF were raised in light - dark photoperiod (12h: 12h) for a week and then were divided into four Mn -exposure groups and four control groups. Mn - exposure groups were intraperitoneally injected with Mn (30mg/kg MnC12o4H20) and the control groups were administered with equal volume of water, respectively at zeitgeber time of ZT2, ZT8, ZT14 and ZT20 for continuous 21 days. The serum estradiol (E2), follicle- stimulating hormone (FSH) and testosterone (T) level was measured by immune chemiluminometric assays ( ICMAS), the sperm quality and quantity was measured too. Results Compared with the corresponding control groups, Mn - exposure groups showed that the sperm quality and quantity, E2, FSH and T level changed, and the results were different at different exposure time. The sperm quantity was decreased in each exposure time, hut sperm activity was decreased only in zeitgeber time of ZT2 and ZT8. The level of estradiol increased only in zeitgeber time of ZT2, and follicle - stimulating hormone decreased in zeitgeber time of ZT8, ZT14 and ZT20. The levels of testosterone reduced in zeitgeber time of ZT2 and ZT20, but elevated in zeitgeber time of ZT14. The average levels of sperm quality and quantity and reproductive hormones in Mn - exposure groups declined, with reduced or increased amplitude and delayed peak time. Conclusion The sperm damage and hormone secretion disorder caused by manganese exposure varied with time. The exposure time must be considered when evaluating the toxicity of manganese.
