摘 要:目的:该文通过模拟光老化皮肤状态,即制造UVA日光照射后损伤的人真皮微血管内皮细胞(HDMEC)模型,研究桃红四物汤对其白细胞介素-1α(IL-1α)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、内皮型一氧化氮合酶(e NOS)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)表达的影响,并以此来阐述桃红四物汤对UVA辐射后受损HDMEC的保护作用。方法:选用大鼠制作不同浓度桃红四物汤血清模型,将HDMEC细胞株分组,制作UVA辐射细胞损伤模型,并给予不同浓度含药血清以及生理盐水作为干预措施,培养并收集上清液备用,采用ELISA法检测IL-1α、IL-6、TNF-α、SOD、CAT指标,用Western Blot法检测e NOS指标。结果:桃红四物汤可显著抑制UVA辐射后受损的HDMEC中IL-1α、IL-6、TNF-α的表达并提高e NOS、SOD、CAT的表达。结论:(1)UVA辐射可以导致HDMEC损伤;(2)桃红四物汤含药血清通过抑制炎症反应,增加血管活性物质和清除自由基来保护UVA辐射下损伤的HDMEC。Objective:To study the effects of Taohong Siwu Decoction on interleukin-1α(IL-1α),interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),endothelial nitric oxide synthase(e NOS),superoxide dismutase(SOD)and catalase(CAT)expression in human dermal microvascular endothelial cell(HDMEC)model after UVA irradiation,to explain the protective effect of Taohong Siwu Decoction on HDMEC damaged by UVA radiation. Methods:In this study,rats were used to prepare different concentrations of Taohong Siwu Decoction,and HDMEC cell lines were divided into groups to prepare UVA radiation cell injury models,and different concentrations of serum containing serum and saline were taken as intervention measures. Supernatants were cultivated and collected as reserves. The levels of IL-1α,IL-6,TNF-α,SOD and CAT were detected by ELISA,and the e NOS was detected by Western Blot. The indexes of IL-1α,IL-6,TNF-α,SOD,CAT were detected by ELISA and the indexes of e NOS were detected by Western Blot. Results:Taohong Siwu Decoction could significantly inhibit the expression of IL-1α,IL-6 and TNF-α in HDMEC induced by UVA irradiation and increase the expression of e NOS,SOD and CAT. Conclusion:(1)UVA radiation can lead to HDMEC damage;(2)Taohong Siwu Decoction serum could protect the damage under UVA radiation HDMEC by inhibiting the inflammatory response,increasing the vasoactive substances and scavenging free radicals.
