摘 要:目的:分析内向整流钾通道2.1蛋白(Kir 2.1)在小鼠伏隔核区中等多棘神经元的表达分布模式。方法:成年Drd1-td Tomato/Drd2-e GFP双转基因小鼠,制备含伏隔核区冠状脑片,运用免疫组织化学方法对Kir 2.1进行染色。通过激光共聚焦显微镜成像及应用Imaris等软件对表达Drd1的多巴胺受体1型(D1)中等多棘神经元(D1-MSNs)和表达Drd2的多巴胺受体2型(D2)中等多棘神经元(D2-MSNs)胞体上Kir 2.1免疫荧光阳性信号的密度进行分析。结果:Kir 2.1阳性信号颗粒在伏隔核区域的分布密度大概为0.3个/μm2。D1-MSNs胞体上Kir 2.1荧光像素值与D2-MSNs相比无显著差异。结论:Kir 2.1在伏隔核区多巴胺能神经元介导的直接通路及间接通路上的表达含量相似。Objective: To analyze the expression pattern of Kir( inwardly rectifying potassium channel) 2. 1 channel protein in medium sized spiny neurons( MSNs) of the nucleus accumbens. Methods: Adult Drdl-td Tomato/Drd2-e GFP double transgenic mice were used for preparing coronal brain slices containing nucleus accumbens. Kir 2. 1 was stained by immunohistochemistry. The immunostaining desnity of Kir 2. 1 in MSNs expressing the Drd1( dopamine receptor type 1,D1-MSNs) and MSNs expressing the Drd2( dopamine receptor type 2,D2-MSNs) was analyzed by laser confocal microscopy and Imaris image processing software. Results: The density of Kir 2. 1-positive signals in the nucleus accumbens was 0. 3 spots/μm2. There was no significant difference in the immunofluorescent pixel value of Kir 2. 1 in the D1-MSNs compared with the D2-MSNs. Conclusion: Kir 2. 1 was similarly expressed in direct and indirect pathways mediated by dopaminergic neurons in the nucleus accumbens.
