摘 要:为研究番木瓜畸形花叶病毒(PLDMV)hc-pro基因保守基序FRNK对病症表现的影响,本研究采用定点突变的方法,获得了PLDMV-DF的hc-pro基因保守基序FRNK的精氨酸(R)突变为异亮氨酸(I)的突变体克隆p35SPLDMV-GFP-R_(183)/I,该位点突变显著减轻了PLDMV在番木瓜上的病症表现,表明是影响其致病性的关键位点。本研究结果为利用交叉保护防控番木瓜畸形花叶病毒病提供了重要参考和材料。To investigate the effect of the hc-pro gene conserved FRNK motif in Papaya leaf distortion mosaic virus (PLDMV), we used the Hainan severe strain PLDMV-DF carrying a green fluorescent protein (gfp) gene as a model virus (denoted as p35S-PLDMV-GFP) to generate a mutant clone of p35S-PLDMV-GFP-Rls3/I, which contained the substitution of Ile for Arg at position 183 in the conserved FRNK motif of PLDMV, which could cause a dramatic symptom change from severe to mild in papaya. The site mutation significantly reduced the expression of PLDMV in papaya, indicating that it is the key point affecting its pathogenicity. The results of this study would provide important references and materials for cross-protection against papaya terato-mosaic virus disease.
