摘 要:目的本研究旨在进一步明确低氧环境运动对老年2型糖尿病大鼠(T2DM)糖代谢、胰岛素敏感性的影响,评估其对糖尿病大鼠骨量丢失和骨质疏松的风险。方法本研究采用72只大鼠55周龄雌性SD大鼠随机分为普通饲料喂养对照组(NC组)和高脂饲料喂养组(HF组),进行8周高脂膳食喂养,通过血糖测试和葡萄糖耐量试验,判断建模是否成功;将建模成功的糖尿病大鼠进行12周的低氧或常氧运动干预,于12周干预结束后进行腹腔动脉取血。结果本研究发现12周干预后常氧糖尿病运动组(NTS组)和低氧糖尿病运动组(LTS组)大鼠体重显著下降,且LTS组显著低于NTS组(P<0.05),LTS大鼠FINS、FPG、HOMA-IR、ISI、AUC_(BG)均显著优于NC组、LC组、NTS组(P<0.05),NTS组和LTS组大鼠骨密度、鲜股骨重、最大应力、最大载荷、断裂载荷、弹性模量均差异无统计学意义(P>0.05)。结论本研究认为低氧环境干预既能够显著改善T2DM大鼠胰岛素敏感性,也能够诱发骨密度下降和骨生物力学性质改变;但在低氧环境下进行中等强度运动能够有效改善老年T2DM大鼠胰岛素抵抗、提高胰岛素敏感性,同时不会降低骨密度、改变骨生物力学性质。Objective To further clarify the effect of exercise in hypoxic environment on glucose metabolism and insulin sensitivity in type 2 diabetic rats and to assess the risk of bone loss and osteoporosis in diabetic rats. Methods Seventy-two 55-week-old female Sprague-Dawley rats were randomly divided into normal feed control group( NC group) and high fat diet fed group( HF group). They were fed for 8 weeks. Serum glucose and glucose tolerance were tested to determine whether the model was successful. The successful model diabetic rats were subjected to hypoxia or normoxia exercise for 12 weeks. The celiac artery blood was taken in 12 weeks after the intervention. Results In the present study,the body weight of rats in NTS group and LTS group decreased significantly after 12 weeks of intervention. The levels of FINS,FPG,HOMA-IR,ISI,and AUCBG in hypoxic group were significantly lower than those in NTS group( P 〈 0. 05). There was no significant difference in bone mineral density,fresh weight,maximum stress,maximum load,breaking load,and elastic modulus between NTS group and LTS group( P 〉 0. 05).Conclusion This study suggests that hypoxic environment intervention significantly improves insulin sensitivity in T2 DM rats and induces decline in bone mineral density and changes in biomechanical properties. However,moderate-intensity exercise in hypoxic environment effectively improves insulin resistance in T2 DM rats,while it does not reduce bone mineral density and change the biomechanical properties.
