摘  要:目的研究槲皮素对高糖刺激的系膜细胞外基质分泌的影响。方法用噻唑蓝比色法(MTT)检测细胞活力和细胞增殖,用流式细胞术检测细胞周期,用Real-time PCR的方法检测P38MAPK、CREB1及TGFβ1的基因表达,用免疫印迹法(Western blotting)检测P38MAPK、CREB1的磷酸化蛋白表达及TGFβ1的蛋白表达,ELISA试剂盒检测上清液FN及ColⅣ蛋白。结果槲皮素能够抑制高糖刺激的系膜细胞的增殖,降低P38MAPK及CREB1的磷酸化水平,下调TGFβ1基因和蛋白表达,减少FN及ColⅣ蛋白分泌。结论槲皮素抑制高糖刺激的系膜细胞P38MAPK信号通路的激活,从而减少细胞外基质的分泌。OBJECTIVE To investigate the influence of quercetin on extracellular matrix secretion of mesangial cells induced by high glucose. METHODS MTT assay was used to observe the cell viability and cell proliferation. Flow cytometry was used to determine the cell cycle. Real-time PCR assay was used to detect the RNA expression levels of P38 MAPK、CREB1 and TGFβ1. Western blotting assay was used to detect the phosphorylation levels of P38 MAPK and CREB1,as well as the protein levels of TGFβ1. ELISA kit was used to detect the FN and Col Ⅳ in the supernatant. RESULTS Quercetin could inhibit cell proliferation,decreased the phosphorylation levels of P38 MAPK and CREB1,downregulated the mRNA and protein levels of TGFβ1,and reduced the secretion of FN and Col Ⅳ in high glucose stimulated mesangial cells. CONCLUSION The study confirmed that quercetin can reduce the extracellular matrix secretion by suppressing the activation of P38 MAPK signal pathway in high glucose stimulated mesangial cells.
