摘 要:目的探讨补阳还五汤在病毒性心肌炎(VMC)心肌纤维化中的作用。方法于小鼠腹腔注射柯萨奇B3病毒液构建病毒性心肌炎模型,分别给予卡托普利混悬液(100 mg·kg^(-1)·d^(-1))和不同浓度的补阳还五汤(18.48,9.24,4.62 g·kg^(-1)·d^(-1))连续干预40 d。分别于给药后第4,10,20,40天每组取6只小鼠,心脏超声检测心功能后,断颈处死,计算心脏体重比,紫外分光光度仪测定羟脯氨酸含量,计算心肌胶原含量。结果与对照组比较,模型组心脏体重比增大,左心室舒张末期内径(LVEDd)、左心室收缩末期内径(LVEDs)增大,缩短分数(FS)和射血分数(EF)下降,心肌胶原含量增多,差异有统计学意义(P<0.05);与模型组比较,补阳还五汤呈剂量依赖性地降低心脏体重比,降低心脏LVEDd、LVEDs,升高FS、EF,减少心肌胶原含量,差异有统计学意义(P<0.05)。结论补阳还五汤可抑制VMC心肌纤维化,改善心功能。OBJECTIVE To study the role of Buyanghuanwu decoction in the pathogenesis of myocardiac fibrosis in viral myocarditis. METHODS The model of viral mycarditis mice was constructed by intraperitoneal injection of Coxsackie B3 virus,respectively,the Captopril suspension( 100 mg·kg^-1·d^-1) and the different concentrations of Buyanghuanwu decoction( 18.48,9.24,4.62 g·kg^-1·d^-1) were given by gavage for 40 days.6 mices every groups were killed after evaluated cardiac function,and then calculated the heart weight ratio,measured hydroxyproline content by ultraviolet spectrophotometry and computed myocardial collagen content on4,10,20 and 40 days after drugs given. RESULTS Compared with the control group,the heart weight ratio of the model group increased,lvedd and lveds increased,FS and EF decreased,and the myocardial collagen content increased,which had statistically significance( P〈0.05); Compared with the model group,Buyanghuanwu decoction showed a dose-dependent for reduced heart weight ratio,decreased LVEDd,LVEDs,increased FS and EF,decreased myocardial collagen content,,which had statistically significance( P〈0.05). CONCLUSION Buyanghuanwu decoction can inhibit viral myocarditis myocardial fibrosis and improve cardiac function.
