磷酸二甲啡烷胶囊在健康人体的药动学研究

摘　　要：目的研究磷酸二甲啡烷胶囊单次及连续多次给药的药代动力学特征,为其临床用药提供可靠依据。方法 12例受试者采用随机、开放、二重3×3拉丁方试验设计,单次及连续多次给药后,研究磷酸二甲啡烷胶囊在健康人体内药代动力学特征。结果单次给药10 mg、20 mg、40 mg后,二甲啡烷主要药动学参数:cmax为(1.430±1.265)μg·L^(-1)、(3.502±2.917)μg·L^(-1)、(5.748±4.635)μg·L^(-1),tmax为(3.13±1.32)h、(2.83±0.58)h、(3.17±1.63)h,AUClast为(15.71±15.38)μg·h·L^(-1)、(34.43±27.97)μg·h·L^(-1)、(70.40±66.48)μg·h·L^(-1),t1/2为(8.75±3.45)h、(9.99±2.99)h、(9.53±2.95)h;多次给药20 mg后二甲啡烷主要药动学参数:cmax为(7.225±6.646)μg·L^(-1)、tmax为(2.38±0.71)h、AUClast为(124.02±129.08)μg·h·L^(-1)、t1/2为(13.24±3.08)h。结论单次给药10~40 mg的人体药动学特征符合一级速率线性动力学过程;20 mg多次给药呈线性动力学累加,体内消除符合一室模型;药动学参数与文献报道基本一致。OBJECTIVE To investigate the pharmacokinetics characteristics of dimemorfan phosphate capsules at single doses or multiple doses in order to provide reliable basis clinical medication. METHODS Single and multiple doses of dimemorfan phosphate capsules（ single dose for 10 mg,20 mg,and 40 mg） were given to 12 healthy volunteers according to an open,random 3 × 3 Latin design. Pharmacokinetics characteristics in healthy human body after the medicine are given. RESULTS The main pharmacokinetic parameters of dimemorfan after single doses dimemorfan phosphate capsules（ 10,20 and 40 mg） were as follows： cmaxwas（ 1. 430 ±1.265） μg·L^-1,（ 3.502±2.917） μg·L^-1 and（ 5.748±4.635） μg·L^-1,tmaxwas（ 3.13±1.32） h,（ 2.83± 0.58） h and（ 3.17± 1.63） h,AUClast was（ 15.71±15.38） μg·h·L^-1,（ 34.43±27.97） μg·h·L^-1 and（ 70.40±66.48） μg·h·L^-1,t1/2 was（ 8.75±3.45） h,（ 9.99±2.99） h and（ 9.53±2.95） h; The main parameters of dimemorfan after multiple doses（ 20 mg） were as follows： cmaxwas（ 7.225±6.646） μg·L^-1,tmaxwas（ 2.38±0.71） h,AUClastwas（ 124.02±129.08） μg·h·L^-1,t1/2 was（ 13.24±3.08） h. CONCLUSION After 10-40 mg single dose,the pharmacokinetic results show that they accord with the process of first order rate linear dynamics. It shows a linear dynamics accumulation after 20 mg multiple doses. Drug elimination in body accords with one-compartment model. The parameter of pharmacokinetics basically coincides with the literature.
