摘 要:[目的]研究黄芪多糖对脓毒症大鼠心脏功能及炎症因子的影响,为中西医结合治疗脓毒症提供理论依据。[方法]55只SD大鼠随机分为对照组、脓毒症组、黄芪多糖(Astraglaus polysaccharide,APS)实验组,脓毒症组与APS实验组按照动物模型建立到处死动物的时间不同设为2、4、6、12、24h共5个时间点亚组。对照组大鼠予腹腔注射0.9%氯化钠溶液作为空白对照,脓毒症组大鼠予腹腔注射大肠杆菌内毒素(lipopolysaccharides,LPS)5mg/kg制作脓毒症模型,APS实验组予注射用APS(200mg/kg·d)干预处理7d后制作脓毒症模型。分别观察大鼠一般情况,心脏病理损害,心室导管法检测大鼠左心室心功能参数,免疫组化检测心室肌细胞肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-1β(interleukin-1β,IL-1β)、IL-6的表达。[结果]脓毒症组大鼠心脏病理损害严重,左心室收缩、舒张功能较对照组下降(P<0.05),TNF-α、IL-1β、IL-6先升高后降低,表达较对照组增强(P<0.05)。APS实验组大鼠一般情况改善,心脏损害减轻,左心室收缩、舒张功能较脓毒症组改善(P<0.05);TNF-α、IL-1β、IL-6先升高后降低,表达较对照组增强(P<0.05),较脓毒症组减弱(P<0.05)。[结论]APS能够改善脓毒症大鼠左心室收缩、舒张功能,抑制脓毒症大鼠心室肌TNF-α、IL-1β、IL-6的表达,减轻脓毒症大鼠心脏损害。[Objective]To study the effect of Astragalus polysaccharides(APS) on cardiac function and inflammatory factors in rats with sepsis, providing theoretical basis for the treatment of sepsis with combination of traditional Chinese and western medicine.[Methods]Fifty-five Sprague Dawley rats were separated into control group, sepsis group and APS experimental group randomly. All the animals in sepsis group and APS experimental group were sacrificed at 2h, 4h, 6h, 12h and 24h from the time of building models. Rats in the control group were injected normal saline intraperitoneally. Rats in the sepsis group were injected lipopolysaccharides from Escherichia coli(5mg/kg) intraperitoneally to make sepsis rats model. Rats in the APS group were preprocessed with APS(200mg/kg·d) before 7 days injecting LPS to make sepsis rats model. At the corresponding time of points, observing the general state in health of the rats, and the hearts were isolated for morphologic pathology. Cardiac functions parameters were detected with a ventricular catheter in the left ventricle of rats,and immunohistochemical observation of ventricular myocyte factors TNF-α,IL-1β and IL-6.[Resuhs]In the sepsis group, the cardiao pathology was severe, the ventricular systolic and diastolic function was decreased compared with control group(P〈0.05), and the TNF-α, IL-1β and IL-6 levels of ventricular myocytes increased first and then decreased, and the expression was increased compared with control group(P〈0.05). In APS experimental group, the general condition of rats was improved, the cardiac damage reduced, the ventricular systolic and diastolic function was improved compared with sepsis(P〈0.05), and the TNF-α,IL-1β and IL-6 levels of ventricular myocytes increased first and then decreased, and the expression was increased compared with control group(P〈0.05), which was decreased compared with sepsis group(P〈0.05).[Conclusion]APS can improve ventricular systolic and systolic function in rats with sepsis, a
