益气活血解毒法对脓毒症大鼠肺保护作用研究

摘　　要：目的：观察不同剂量化纤胶囊对脓毒症大鼠死亡率、血气分析及进行肺组织匀浆MDA的测定、SOD活力测定,明确益气活血解毒法化纤胶囊对脓毒症大鼠肺的保护作用。方法：通过对大鼠盲肠结扎穿刺制备脓毒症实验模型。选取SPF级Wistar大鼠130只,雌雄各半,随机将实验动物分成7组,分别为：假手术组,模型组,生理盐水组,血必净组,化纤胶囊低、中、高剂量组。盲肠结扎手术（CLP）术后2、12 h分别给予低、中、高剂量化纤胶囊组灌胃给药（50、150、200 mg/kg）,生理盐水组给予同样体积0.9%生理盐水,血必净组给予血必净10 mL/kg灌胃给药。模型制备后6、12、24 h各时间点处死动物,采集血液及肺组织进行血气分析、MDA测定和SOD活力测定。结果：化纤胶囊低、中、高剂量组48 h死亡率分别为75%、65%、50%,比模型组及生理盐水组明显降低（P〈0.05）。术后6、12、24 h化纤胶囊低、中、高剂量组大鼠血气分析PaO2、PaO2/FiO2比模型组及生理盐水组显著改善（P〈0.01）,且高剂量组改善明显（P〈0.05）。MDA活性在术后各时间段化纤胶囊低、中、高剂量组比模型组、生理盐水组显著降低（P〈0.01）,且高剂量组降低明显（P〈0.05）。SOD活力化纤胶囊低、中、高剂量组在术后各时间段比模型组及生理盐水组显著升高（P〈0.01）,且高剂量组差异明显（P〈0.05）。结论：益气活血解毒法化纤胶囊对脓毒症大鼠肺损伤有保护作用,并在一定程度上提高大鼠生存率。Objective ：To observe the effect of different doses of Huaxian Capsule on rats with sepsis mortality,blood gas analysis and lung homogenate MDA assay,SOD activity assay,to make a clear protective effect of Yiqi Huoxue Jiedu method of Huaxian Capsule on sepsis rats lung. Methods ：The experimental model of sepsis was prepared by cecal ligation and puncture in rats. Selected the SPF 130 Wistar rats,male and female,were randomly divided into 7 experimental animal groups,respectively ：sham operation group,model group,saline group,Xuebijing group,Huaxian Capsule low,middle and high dose group. Cecal ligation surgery（CLP）after 2 h,12 h were given low,medium and high dose Huaxian Capsule group by gavage（50 mg/kg,150 mg/kg,200 mg/kg）,normal saline group were given the same volume of 0.9% saline,xuebijing group were given intragastric administration of Xuebijing 10 mL/kg. After the model preparation,6 h,12 h and 24 h were killed at each time point. Blood and lung tissues were collected for blood gas analysis,MDA assay and SOD activity determination. Results ：The mortality of 48 h in the low,medium and high dose groups of Huaxian Capsule was 75%,65% and 50% respectively,which was lower than that of model group and normal saline group（P〈0.05）. The blood gas analysis,PaO2 and PaO2/FiO2 in the low,medium and high dose groups of rats with 6 h,12 h and 24 h were significantly improved（P〈0.01）compared with the model group and the saline group,and the high dose decreased significantly（P〈0.05）.The MDA activity in the Huaxian Capsule group was significantly lower than that in the model group and saline group at the different time after operation（P〈0.01）,and the high dose group decreased obviously（P〈0.05）. SOD dynamic Huaxian Capsule group was significantly higher than the model group and saline group at each time after operation（P〈0.01）,and the difference was significant among the high dose group（P〈0.05）.
