摘 要:目的:探讨精简平喘方不同制备方法干预对哮喘小鼠免疫功能的影响。方法:以10%OVA/AL(OH)3致敏、5%OVA激发制成支气管哮喘小鼠模型,以精简平喘方的两种不同提取方法所得提取物为干预手段,于激发前开始给药,至激发结束取材。观察肺组织病理、外周血及BALF中EOS、Ig E,以及肺组织中IL-4、IFN-γ水平。结果:中药A(含挥发油)、B对哮喘模型小鼠EOS及总Ig E、肺组织IFN-γ、IL-4均有较为明显的干预作用,能够显著降低EOS、Ig E、IL-4,升高IFN-γ(均P〈0.05)。其中,低、中剂量中药A组对外周血EOS的降低作用优于同剂量中药B组(均P〈0.05),中药A高剂量优于低、中剂量组(均P〈0.05);中、高剂量中药A组对BALF中EOS的降低作用优于同剂量中药B组(均P〈0.05),中药A高剂量优于低、中剂量组(均P〈0.05);高剂量中药A组对外周血中Ig E的降低作用优于同剂量中药B组(P〈0.05),中药A高剂量优于低、中剂量组(均P〈0.05);中、高剂量中药A组对BALF中Ig E的降低作用优于同剂量中药B组(均P〈0.05),中药A高剂量优于低、中剂量组(均P〈0.05);中、高剂量中药A组对肺组织中IFN-γ的降低作用和IL-4的升高作用均优于同剂量中药B组(均P〈0.05),且优于中药A低、中剂量组(均P〈0.05)。结论:中药A、B对哮喘模型小鼠EOS及总Ig E、IFN-γ、IL-4均有较为明显的干预作用,能够显著调节上述指标,调节哮喘小鼠的免疫失衡。其中,中药A的干预效果总体优于中药B,以中、高剂量组优势更为突出。Objective :To investigate the effect of different preparation methods of Simplified PingchuanDecoction on immune function in asthmatic mice. Methods :Asthmatic mice model were established by 10% OVA/AL(OH)3 sensitization and 5% OVA stimulation. Extracs from Pingchuan Decoction by two different preparation methods of low,mediu,and high dosage were administered before the stimulation and finished at the end of the excitation. The lung pathology,total Ig E,EOS in peripheral blood and BALF,and the levels of IL-4 and IFN-γ in lung tissue were observed. Results :Both of the groups of Chinese medicine A(including volatile oil)and B can significantly reduce the EOS,Ig E,IL-4 levels,and increase IFN-γ(P〈0.05). Peripheral blood EOS reduction in the low and medium dosage groups of Chinese medicine A was superior to the same dosage group of Chinese medicine B(P〈0.05),Chinese medicine A high dosage had better effect than low and medium dosage group(P〈0.05). The effect of medium and high dosage groups Chinese medicine A on the reduction of EOS in BALF were better than that of the same dosage of traditional Chinese medicine B group(P〈0.05),and the high dosage group of Chinese medicine A was better than of low and middle dosage groups(P〈0.05). High dosage group of Chinese medicine A was superior to the same dose of traditional Chinese medicine B group(P〈0.05)in the reduction of Ig E in peripheral blood,and the high dosage group of Chinese medicine A was better than the low and medium dosage groups(P〈0.05). The effect of medium and high dosage groups of Chinese medicine A on the reduction of Ig E in BALF was better than that of the same dosage groups of Chinese medicine B(P〈0.05),and the high dose group of Chinese medicine A were better than that of low and middle dose group(P〈0.05). Medium and high dosage groups of Chinese medicine A had better effect on decrease of IFN-γ and increase of IL-4 levels in lung tissue than the same dosage group of Chinese medicin
