芪山无糖颗粒对糖尿病前期大鼠瘦素和肿瘤坏死因子-α表达的影响

摘  要:目的观察芪山无糖颗粒剂对糖尿病前期大鼠慢性炎症因子瘦素和肿瘤坏死因子-α表达水平的变化。方法采用SD雄性大鼠,通过高脂高糖饲料喂食制造糖尿病前期模型。然后将模型大鼠分为四个干预组,分别以小剂量(Z1组)、中剂量(Z2组)、大剂量(Z3组)中药芪山无糖颗粒剂和罗格列酮(X)进行干预,同时设立普通饲料喂食的正常对照组(NC)和单独高糖高脂饲料喂食的模型组(MC)。干预16周后,测量体重;以全自动生化仪检测血糖(部分采用试纸法)、血脂;采用ELISA检测肿瘤坏死因子-α(TNF-α)、瘦素(Leptin)、血清胰岛素。结果与NC组比较,MC组Leptin和TNF-α明显升高(P〈001);与模型组比较,药物组出现一定程度的恢复(P〈0.05或0.01).Z2、Z3、X组下降显著(P〈0.01)。与MC组比较,药物干预各组均出现血糖、血脂降低,Z3、X组血糖、血脂降低更明显(P〈0.01)。与NC组比较,MC组胰岛素水平明显增加(P〈0.01);与增高的MC组比较,Z3、X组两者胰岛素降低最明显(P〈0.01),而Z3与X组之间差异无统计学意义(P〉0.05)。结论芪山无糖颗粒明显降低糖尿痛前期大鼠TNF-α和leptin水平,改善糖尿病前期大鼠糖代谢和脂代谢,可缓解糖尿病前期大鼠高胰岛素血症,降低胰岛素抵抗程度。Objective To observe the effects of Qishan granules without sugar on inflammatory factors leptin and TNF-α expression in the prediabetes rats. Methods White rat models with prediabetes were first established through feeding themselves on high-fat diets. Qishan granules without sugar were used as intervention treatment, the CTM groups were divided into small-dose group ( Z1 ) , medium-dose group ( Z2 ) and high-dose group ( Z3 ) , and rosiglitazone in the positive contract groups (X) . After 10 weeks, the rat models' weight, FBG, 2hPG, FLNS, 2hlNS, leptin, and TNF- ct were tested. Results Compared with control groups, Leptin and TNF-α obvious were increased in model groups ( P〈0.01 ) . Compared with model groups, the levels of Leptin and TNF-α were decreased in the drugs groups (P〈0.05或0.01) , and significant decreased in Z2, Z3 and X groups. Compared with model groups, the levels of blood glucose and blood fat were all decreased in the drugs groups, and significant decreased in Z3 and X groups ( P〈0.01 ) . Compared with the control groups, the levels of insulin were obvious increased in the model groups. Compared with model groups, the levels of insulin were distincdy decreased in Z3 and X groups, but there were no marked differences between the two groups (P〉0.05) . Conclusion Qishan granules without sugar remarkably improves the weight, metabolism of glucose and lipid of the white rat models in the pre- diabetes, increases the sensitivity of insulin and also decreases the concentration ofleptin and TNF-α in the blood.
