摘 要:目的:分析在气道模型重塑的幼龄哮喘大鼠模型中,健脾补肺化痰方对大鼠肺功能、肺组织形态学参数及PDGF和ERK1/2/c-jun变化的影响。方法:将40只清洁级幼龄大鼠随机分为正常组、模型组、健脾补肺化痰方组和地塞米松组并建立哮喘气道重塑模型。测定肺功能相关指标并通过肺组织切片HE染色,分析大鼠肺组织形态学变化。使用逆转录聚合酶链反应(RT-PCR)和免疫印迹法(Western-blot)测定PDGF、ERK1/2和c-jun表达变化。并采用直线相关法性分析PDGF、ERK1/2和c-jun与大鼠气道形态学参数的相关性。结果:与哮喘模型组比,健脾补肺化痰方组大鼠呼吸症状和支气管炎性细胞浸润较轻,大鼠PEF、FEV0.2、FEV0.2/FVC及肺组织中PDGF、ERK1/2、c-junm RNA表达显著升高(P<0.05)。气道形态学测量结果显示模型组大鼠WAt/Pbm、WAi/Pbm及WAm/Pbm较其他各组显著增加(P<0.05)。PDGF、ERK1/2及c-jun蛋白的表达水平与WAt/Pbm、WAi/Pbm及Wam/Pbm呈正相关关系(P<0.01)。结论:健脾补肺化痰方能够抑制炎症反应,改善肺功能及气道形态学参数,通过PDGF、ERK1/2/c-jun信号转导通路实现对气道重塑的抑制。Objective: To analyze the effect of Jianpi Bufei Huatan decoction on pulmonary function, lung morphologi- cal parameters and PDGF and ERK1/2/c-jun changes in airway remodeling young asthma rats. Method: 40 young SD rats were randomly divided into normal group, model group, Jianpi Bufei Huatan decoction group and dexamethasone group, using ovalbumin(OVA) atomization inspired rat model of asthma. Pulmonary function parameters were measured and his- tological changes of lung tissue were analyzed by HE staining. Using RT-PCR and Western blotting to detect the change of PDGF and ERK1/2/c-jun. The linear correlation analysis was used to analyze the correlation between PDGF, ERK1/2, c-jun and airway morphological parameters in rats. Results: Compared with the model group, the respiratory symptoms and bronchial inflammatory cell infiltration of the rats in the Jianpi Bufei Huatan decoction group were lighter, and the expression of PDGF, ERK1/2, and c-jun mRNA in rat was significantly increased(P〈0.05). The results of airway mor- phology showed that the rats in the model group had significantly higher WAt/Pbm, WAi/Pbm, and WAm/Pbm than other groups(P〈0.05). The expression levels of PDGF, ERK1/2, and c-jun were positively correlated with WAt/Pbm, WAi/Pbm and Wam/Pbm(P〈0.01). Conclusion: Jianpi Bufei Huatan decoction can inhibit inflammation, improve pulmonary function and inhibit airway remodeling through PDGF and ERK1/2/c-jun signal transduction pathwavs.
