摘  要:目的:观察电针对围绝经期抑郁症大鼠Wnt/β-catenin信号通路的影响,探讨电针治疗抑郁的作用机制。方法:将雌性SD大鼠按照随机数字表法分为空白组、模型组、药物组和电针组,每组12只;除空白组外,其余大鼠采用双侧卵巢切除术和慢性不可预见刺激性应激法制备围绝经期抑郁症大鼠模型,用悬尾实验评价大鼠的行为学改变,测量海马组织中雌二醇(E2)和去甲肾上腺素(NE)的含量,分析围绝经期抑郁情况。药物组采用盐酸氯米帕明灌胃,电针组采用电针治疗“百会”、“三阴交”和“肾俞”。采用酶联免疫吸附法(ELISA法)检测大鼠海马组织中Dickkopf-1(DKK1)、低密度脂蛋白受体相关蛋白-5(LRP-5)和低密度脂蛋白受体相关蛋白-6(LRP-6)的表达情况,分析其表达与围绝经期抑郁的相关性。结果:模型组大鼠海马组织E2和NE含量降低,悬尾不动时间明显增加,与空白组比较,差异有统计学意义(P〈0.05);模型组海马组织DKK1蛋白表达下降,LRP-5和LRP-6蛋白表达升高。经过28 d治疗,药物组和电针组E2和NE含量升高,DKK1表达升高,LRP-5和LRP-6表达下降,与模型组比较,差异有统计学意义(P〈0.05)。结论:电针治疗通过调节海马中DKK1、LRP-5和LRP-6蛋白的表达影响Wnt/β-catenin信号通路,促进海马神经细胞的修复,减少细胞调亡,起到抗抑郁作用。Objective: To observe the effects of electroacupuncture( EA) on Wnt/beta-catenin signaling pathway in the hippocampus of perimenopausal depression rats so as to explore the mechanism of EA treatment of depression. Methods: According to random number table method,SD female rats were randomly divided into blank group,model group,drug group and EA group,with 12 rats in each group. Except the blank group,the rest of rats were established with bilateral overiectomy and chronic unpredictable mild stress( CUMS) to simulate perimenopausal depression model. By using the rat tail suspension test,we evaluated behavioral change,E2(Estradiol) and NE(Norepinephrine) in the hippocampus to judge the depression. Medicine group was treated with clomipramine hydrochloride orally and EA group received EA therapy at Baihui( DU20),Sanyinjiao( SP6) and Shenshu( BL23). After the success of the building,we detected Dickkopf 1( DKK1),low density lipoprotein receptor related protein-5( LRP-5) and low density lipoprotein receptor related protein-6( LRP-6) in the hippocampus by immunoassay( ELISA) to analyze its expression and correlation of perimenopausal depression. Results: In the model group,E2 and NE reduced and the tail suspension motionless time increased significantly. Compared with the blank group,the difference was statistically significant( P〈 0. 05). In the model group,DKK1 decreased,LRP-5 and LRP-6 increased in the hippocampus. After 28 days treatment,drug group and EA group's E2 and NE increased,DKK1 expression increased,LRP-5 and LRP-6 expression decreased. Compared with model group,the difference was statistically significant( P〈0. 05). Conclusion: EA treatment affects the Wnt/beta-catenin signaling pathway by adjusting the expressions of DKK1,LRP-5 and LRP-6 in the hippocampus,promotes hippocampal neural stem cells proliferation and reduces cell apoptosis to have antidepressant effect.
