摘 要:目的:建立加味升降散中5种大黄蒽醌类成分(芦荟大黄素、大黄酸、大黄酚、大黄素、大黄素甲醚)含量的测定方法。方法:YMC-Pack ODS-AM C18色谱柱(250 mm×4.6 mm,5μm),柱温:25℃;以甲醇为流动相A,0.1%磷酸溶液为流动相B,梯度洗脱,检测波长254 nm,流速1.0 m L·min-1。结果:芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚分别在0.257 42.059 0,0.163 51.308 2,0.098 20.785 7,0.415 13.321 0,0.080 40.643 1μg范围内线性关系良好(r≥0.999 8),平均加样回收率(n=9)分别为97.01%(RSD 1.28%),99.67%(RSD 2.17%),98.97%(RSD 2.3%),99.52%(RSD 1.95%),99.04%(RSD 1.82%)。结论:实验中建立的研究方法重现性好、操作简便、准确可靠,可应用于加味升降散水煎剂的质量控制。Objective: To establish the determination method of the five Rhubarb anthraquinones constituents in Shengjiang Powder which contained the aloe - emodin, rhein, chrysophanol, emodin and rheochrysidin. Methods : The a- nalysis was achieved on a YMC -Pack ODS -AM Cls column (250 mm×4.6 mm,5 μm) with the mobile phase of meth- anol(A) -0.1% phosphate acid(B) ,at a flow rate of 1.0 mL · min-1. The detection was at 254 nm and temperature was at 25 ~C. Results : The liner rangers of aloe - emodin, rhein, emodin, chrysophanol and rheochrysidin were within 0. 257 4 - 2.059 0 μg( r = 0.989 9 ) ,0. 163 5 - 1. 308 2 μg( r = 0. 998 ) ,0. 098 2 - 0.785 7 μg( r = 0. 999) ,0.415 1 - 3. 321 0 μg( r = 0. 999 9 ) , 0. 080 4 - 0. 643 1 μg ( r = 0. 999 8 ). The average recovery rates were 97.01% ( RSD 1.28% ) ,99.67% ( RSD 2.17% ) ,98.97% ( RSD 2.33% ) ,99.52% ( RSD 1.95% ) ,99.04% ( RSD 1.82% ), respec- tively( n = 9). Conclusion: This method is easy,reproducible, accurate and reliable,which could be used for quality con- trol of Shengiiang Powder.
