镇肝熄风汤对自发性高血压大鼠主动脉弓血管平滑肌细胞凋亡的影响  

Effect of Zhengan Xifeng Decoction on Apoptosis of Aortic Arch Vascular Smooth Muscle Cells in Spontaneously Hypertensive Rats

摘　　要：目的：研究不同剂量镇肝熄风汤对自发性高血压大鼠（SHR）主动脉弓血管平滑肌细胞凋亡的影响。方法：20周龄自发性高血压大鼠（SHR），雄性，12只，模型组、中药低剂量组、中药中剂量组、中药高剂量组、复方罗布麻组。每组12只，同源雄性京都威斯特大（WKY）12只作为对照组。灌胃给药45d后，采用TUNEL标本制备。检测法测定大鼠主动脉弓血管平滑肌细胞凋亡。结果：模型组中出现大量血管平滑肌细胞凋亡，中药中剂量组、中药高剂量组、复方罗布麻组血管平滑肌细胞凋亡明显减少（P〈0．01）。结论：镇肝熄风汤可以有效减少高血压对血管平滑肌细胞凋亡的影响，有效的抑制血管平滑肌细胞凋亡，起到良好的降低血压和保护血管平滑肌细胞的作用．减轻主动脉弓血管重构。Objective ：To study different doses effect of Zhengan Xifeng Decoction on apoptosis of aortic arch vascular smooth muscle cells in spontaneously hypertensive rats （SHR）. Methods：A total of 60 male SHR, 20 weeks old,were ran- domly divided into the model group, low dose, medium dose, high dose Zhengan Xifeng Decoction groups and compound apocynum group, 12 in each group. Twelve homologous male rats （WKY） served as the normal control group. After ga- vage for 45 days, we used the TUNEL method to measure the apoptosis of aortic arch vascular smooth muscle cells in spontaneously hypertensive rats （SHR）. Results：A large number of aortic arch smooth muscle cell apoptosis was in model group. Zhengan Xifeng Decoction medium dose and high dose groups and compound apocynum groups＇ aortic arch vascu- lar smooth muscle cell apoptosis decreased significantly（ P 〈 0. 01 ）. Conclusion：Zhengan Xifeng Decoction can effectively reduce the effect of hypertension on apoptosis of aortic archvascular smooth muscle cells, effectively inhibit the apoptosis of aortic archvascular smooth muscle cells. Zhengan Xifeng Decoction has the effect of lowering blood pressure, protecting the smooth muscle cells of the blood vessel and reducing the remodeling of the aortic archvascular.
