从分子水平探讨益气活血方对肾间质纤维化的干预

摘　　要：目的：从分子水平探讨益气活血方对肾间质纤维化的干预。方法：雄性SD大鼠100只随机分为6组,假手术组10只,模型组10只,血管紧张素转化酶抑制剂（ACEI）组20只,中药低、中、高剂量组各20只。采用单侧输尿管梗阻（UUO）制备肾间质纤维化大鼠模型。假手术组、模型组均给予等剂量生理盐水灌胃,ACEI组给予稀释成0.1 mg/mL的洛汀新2 mL灌胃,中药低剂量组给予0.25 g/mL制剂2 mL灌胃,中药中剂量组给予0.5g/mL制剂2 mL灌胃,中药高剂量组给予1.0 g/mL制剂2 mL灌胃。各组均连续灌胃14 d。结果：与假手术组比较,其余各组BUN和Scr含量,NF-κB及Ⅰ、Ⅲ、Ⅳ型胶原蛋白表达升高,而TGF-β1、TIMP-1、MMP1、MMP2和MMP9 mRNA表达降低（P〈0.05）;与模型组比较,ACEI组、低剂量组、中剂量组和高剂量组BUN和Scr含量,NF-κB及Ⅰ、Ⅲ、Ⅳ型胶原蛋白表达降低,而TGF-β1、TIMP-1、MMP1、MMP2和MMP9 mRNA表达升高（P〈0.05）;与ACEI组比较,中剂量组和高剂量组BUN和Scr含量,NF-κB及Ⅰ、Ⅲ、Ⅳ型胶原蛋白表达降低,而TGF-β1、TIMP-1、MMP1、MMP2和MMP9 mRNA表达升高（P〈0.05）;与低剂量组比较,中剂量组和高剂量组BUN和Scr含量降低,而TGF-β1、TIMP-1、MMP1、MMP2和MMP9 mRNA表达升高（P〈0.05）;中剂量组和高剂量组BUN和Scr含量,NF-κB及Ⅰ、Ⅲ、Ⅳ型胶原蛋白表达,TGF-β1、TIMP-1、MMP1、MMP2和MMP9 mRNA表达比较无统计学差异（P〉0.05）。结论：益气活血方可改善肾间质纤维化大鼠肾功能,降低NF-κB及Ⅰ、Ⅲ、Ⅳ型胶原蛋白表达,增加TGF-β1、TIMP-1、MMP1、MMP2和MMP9 mRNA表达,具有重要研究意义。Objective： To explore the intervention of Yiqi Huoxue Recipe on renal interstitial fibrosis from molecular level. Methods： A hundred male SD rats were randomly divided into 6 groups,10 in the sham operation group,10 in the model group,20 in the angiotensin converting enzyme inhibitor（ ACEI） group and 20 in the low,medium and high dose groups of Chinese medicine. A rat model of renal interstitial fibrosis was prepared by unilateral ureteral obstruction（UUO）. The sham operation group and model group were given the same dose of normal saline. ACEI group received Lotensin diluted into 0. 1 mg/mL 2 mL preparation orally,Chinese medicine low dose group was given 0. 25 g/mL preparation 2 mL orally and Chinese medicine middle dose group received 0. 5 g/mL preparation 2 mL orally,Chinese medicine high dose group was given 1 g/mL preparation 2 mL gavage. Each group was given the medicine 14 d continuously. Result： Compared with the sham group,the other groups＇ BUN and the contents of Scr,NF-κB,Ⅰ,Ⅲ and Ⅳ collagen expression increased,while TGF-β1,TIMP-1,MMP1,MMP2 and MMP9 mRNA decreased（ P〈 0. 05）. Compared with the model group,ACEI group,low,middle and high dose groups＇ BUN and Scr,NF-κB,Ⅰ,Ⅲ and Ⅳ collagen expression decreased,while TGF-β1,TIMP-1,MMP1,MMP2 and MMP9 mRNA increased（ P 〈0. 05）. Compared with ACEI group,middle and high dose groups＇ BUN and the contents of Scr,NF-κB,Ⅰ,Ⅲ and Ⅳ collagen expression decreased,while TGF-β1,TIMP-1,MMP1,MMP2 and MMP9 mRNA increased（ P 〈0. 05）. Compared with the low dose group,middle and high dose groups＇ BUN and the contents of Scr,TGF-β1,TIMP-1,MMP1,MMP2 and MMP9 mRNA increased（ P 0. 05）. In middle and high dose groups,the expressions of BUN,Scr,NF-κB,I,III,IV,B,TGF-β1,TIMP-1,MMP1,MMP2 and MMP9 mRNA were not statistically（ P 〈0. 05）. Conclusion： Yiqi Huoxue Recipe can improve the renal interstitial fibrosis of rats renal function,reduce NF-κB,Ⅰ,Ⅲ and Ⅳ type collagen,increase the expressions of TGF-�
