摘  要:目的:增强凝胶在支持物上的粘附能力,防止彗星电泳时脱胶。方法:Jurkat细胞株与凝胶液混和,涂抹专用检测载玻片和刻痕有机玻璃片,然后进行常规彗星凝胶电泳,荧光显微镜下分析DNA损伤。结果:刻痕有机玻璃片防脱胶率96.8%,与专用试剂盒载玻片没有统计学差异(P>0.05),刻痕有机玻璃片不影响电泳效果。结论:刻痕有机玻璃可用来进行彗星电泳,检测细胞DNA损伤。Objective: To enhance the adhesion of gelatin on support and prevent degumming during comet electrophoresis. Methods: The Jurkat cell line was mixed with the gel,and the mixture was transferred to the special slide glass and the nicking plexiglass. Then,the conventional comet gel electrophoresis was carried out,and the DNA damage was analyzed under the fluorescence microscope. Results: The antidegumming rate of the nicking plexiglas was 96.8%,and there was no statistical difference from that of the special slide glass( P 〉 0. 05), and the nicking plexiglas did not affect the electrophoretic effect.Conclusion: Nicked plexiglass can be used for comet electrophoresis to detect DNA damage in cells.
