摘  要:目的:探讨绞股蓝皂苷(GP)对晚期糖基化终末产物(AGEs)诱导的人肾小球系膜细胞(human glomerular mesangial cells,HMCs)TGF-β1及PDGF表达的抑制作用。方法:采用AGEs诱导HMCs细胞TGF-β1及PDGF表达,采用不同剂量GP细胞72 h后,RT-PCR和Western blotting检测TGF-β1、PDGF的表达。结果:AGEs能诱导HMCs中TGF-β1、PDGF表达。GP对诱导TGF-β1、PDGF表达有抑制作用,呈浓度越高,抑制作用越强。结论:GP可通过抑制TGF-β1、PDGF表达改善糖尿病肾病病情。Objective: To investigate the inhibitory effect of Gynostemma Pentaphyllum( GP) on the expression of human glomerular mesangial cells( HMCs) TGF-beta 1 and PDGF induced by advanced glycation end product( AGEs). Methods: The expression of TGF-beta 1 and PDGF in HMCs cells was induced by AGEs. After 72 h incubation with different doses of GP,RT-PCR and Western blotting were used to detect the expression of TGF-beta 1 and PDGF. Results: AGEs can induce the expression of TGFbeta 1 and PDGF in HMCs. GP inhibited the expression of TGF-beta 1 and PDGF. The higher the concentration,the stronger the inhibition was. Conclusion: GP can improve the condition of diabetic nephropathy by inhibiting the expression of TGF-beta 1 and PDGF.
