摘 要:为了解田间猪繁殖与呼吸综合征病毒(PRRsv)的流行及变异情况,本研究将PRRSV阳性样品接种到Marc-145细胞并传代后成功分离到1株PRRSV,将其命名为HENZMD-9,并对其全基因序列进行测定和分析。结果显示,HENZMD-9为1株类美国NADC30毒株,并与国内高致病性PRRSV(HP—PRRRSV)毒株的2个片段发生重组,这2个片段分别来自HP—PRRSV JXA1株的细胞传代致弱株JXA1-P80及JXA1P45,分别位于基因组5’端及开放式阅读框lab区域,表明HENZMD-9很有可能与JXA1-R疫苗回复株发生了重组,提示不同类型毒株在田间的共存会加剧PRRSV的演变,从而增加临床防控难度。因此,持续监测PRRSV田间流行及变异动态对PRRSV的防控具有一定的,临床指导意义。To understand the prevalence and molecular variation characteristics of porcine reproduc tive and respiratory syndrome virus (PRRSV)in the field, a PRRSV positive sample was isolated and inoculated onto the Marc-145 cell layers and developed by serial passage,then a strain was ob- tained neamed HENZMD-9. The whole genome was then sequenced and analyzed, which showed that HENZMD-9 was a NADC30-1ike strain recombined with highly pathogenic PRRSV (HP- PRRRSV). There existed two substitution fragments which were from the attenuated strains JXA1-PS0 and JXA1-P45 developed by serial passage of HP-PRRSV strain JXA1-R in Marc-145 cells,and were located in the 5rend and ORFlab of the genome,respectively. These results indica- ted that HENZMD 9 was most probably recombined with the revertants of the vaccine strain JXA1-R,warning that coexistence of different kinds of PRRSVs in the fields can aggravate the evolution of PRRSV and thus increase the difficulties in clinical prevention and control. Therefore, continuously monitoring the prevalence and dynamic variation of predominant PRRSVs in the fields has certain clinically guiding significance on PRRSV prevention and control.
