摘 要:为了探讨白头翁素(anemonin)在仔免大肠杆菌性腹泻中的作用及其对核因子NF-κB p65 表达的影响,试验将200只30日龄健康新西兰兔随机分为4组,大肠杆菌感染致腹泻(E.coli)组、E.coli+anemonin干预组、生理盐水(NS)组和NS+anemonin对照组。观察各组仔兔临床症状,作为仔免是否发病的指标,同时统计各组仔兔体质量变化及存活时间,观察肠道组织病理学变化,并用实时荧光定量RT—PCR和Western blot方法分别检测肠道组织内NF-κB p65 基因及其蛋白表达量。结果显示,E.coli组仔兔表现精神沉郁,食欲减退或废绝,排出水样或胶冻样稀粪,脱水,且体质量下降明显;剖检发现肠黏膜充血,水肿;组织学变化显示肠黏膜绒毛排列紊乱,萎缩、断裂、脱落、间隙增宽、空泡化等。而与E.coli组相比,E.coli+anemonin干预组仔免症状则较轻,死亡率明显降低(P〈0.05),感染仔兔存活时间明显延长(P〈0.05);Western blot和qRT—PCR结果显示,anemonin干预可使仔兔肠道组织NF-κB p65 mRNA及其蛋白表达量极显著减弱(P〈0.01),干预后14d恢复至正常表达水平。以上结果表明,E.coil感染诱导仔兔腹泻导致肠损伤过程中可激活NF-κB p65 mRNA及其蛋白表达,这可能是导致仔兔肠损伤的重要因素,而其抑制剂anemonin在干预E.coli致仔兔腹泻引起的肠损伤方面有重要作用。To investigate the effect of anemonin on NF-κB p65 expression in diarrhea stress intestinal mucosa injury induced by Escherichia coli (E. coli) infection in weanling rabbit. A total of 200 New zealand rabbits were randomized into four groups. E. coli group,E, coli + anemonin group, normal saline (NS) group and NS+ anemonin group. The survival time after infection,the intestine histopathology and the mRNA and protein expression of NF-κB in the intestinal tissue were observed at 2,4,6,8, and 14 days after infection. The results showed that histopathologically, the intestinal villus were somewhat random in arrangement,wastage, snapped, fall off and neutrophils infiltrating in E. coli-infected rabbit, with depression, diarrhea and weight loss dramatically; Compared to that of E. coli-infeeted rabbit, the anemonin treatment alleviated clinical signs, prolonged survival time and decreased mortality(P〈0.05);anemonin treatment also markedly down-regulated the levels of mRNA and protein of NF-κB p65 in the intestinal tissue of Escherichia coliinfected rabbits(P〈0.01). On day 14,the expression of NF-κB p65 mRNA and protein restored to normal level. These data demonstrated that NF-κB signaling pathway participated in diarrhea stress intestinal tissue injury probably induced by Escherichia coil-infected in rabbit and anemonin remarkably down-regulated the levels of mRNA and protein of NF-κB p65 during bacterial infection, thus supporting the use of anemonin for managing diarrhea cases of the weaned rabbit.
