气虚体质大鼠模型的实验研究

摘　　要：目的探索气虚体质动物模型的造模方法及其效果，以期为中医体质动物建模量化评估提供参考依据。方法选择SPF级SD雄性大鼠16只，随机分为气虚组和对照组，每组各8只，适应性喂养后对气虚组进行饮食控制和睡眠剥夺干预，通过观察体表特征，测量体重、体温、摄食量、饮水量、抓力、血流变和血常规指标，评估气虚体质大鼠的造模效果。结果气虚体质大鼠体质特征明显，体重减轻，体温变化不明显；摄食、饮水量在造模14d后逐渐减少；抓力显著降低；高、中、低切全血黏度显增高，红细胞显著增多，白细胞显著减少。结论饮食控制和睡眠剥夺组成的造模方案能够造成大鼠机体生理性功能失调。模型大鼠气虚体质体征明显，机体免疫力减弱，力量及抗疲劳能力降低，血液运行不畅，均符合中医气虚体质特征。Objective To explore the method and effect of establishing animal model of qi deficiency constitution in order to provide the reference for the further study of TCM constitution. Methods 16 male SPF SD rats were randomly divided into qi deficiency and control group. After adaptive feeding, the qi deficiency group rats were controlled of diet and deprived of sleep, then to evaluate the effect of qi deficiency constitution model by observing the surface characteristics and measuring the body temperature, body weight, food intake, water intake, grip strength, blood rheology and blood routine index. Results The surface characteristics of qi deficiency constitution rats was obvious with weight loss, but temperature had no change; the intake of food and water decreased in 14 days after modeling; grip strength decreased significantly; the red cells and blood theology indexes increased significantly, but the white blood cells significantly went down. Conclusion The modeling scheme of diet control and sleep deprivation can cause physiological function disorder. The physical signs ofqi deficiency in the model rats were obvious, and the immunity weakened, strength and ability of anti-fatigue decreased, and the blood circulation is unsmooth, which all accord with the physical characteristics of qi deficiency constitution.
