摘  要:本实验以剑尾鱼为试验动物,研究苯并(a)芘在不同药物剂量及作用时间下对CYP1A1基因表达的影响。采用活体实验和体外实验,活体实验,B(a)P浓度500ug·L-1,24h后取样,研究CYP1A1基因在剑尾鱼各组织的表达情况;体外实验,B(a)P浓度为10-10000ug·L-1,分别于给药6.48h后取样,采用实时定量RT-PCR方法对剑尾鱼CYP1A1基因的表达进行了研究。结果表明,CYPlAl在鳃、脑、脾、肝脏、肾、肠中均有表达,在肝脏中的表达量最高;CYP1A1基因的表达与Bfa)P的暴露浓度呈现很好的剂量一效应关系,且10ug·L-1B(a)P的暴露就能显著诱导肝细胞CYP1A1的基因表达(p〈0.05);肝组织的活性会随着时间的延长而降低,高剂量B(a)P诱导下,CYP1A1的表达量相比于同时间段低剂量诱导表达量存在极显著差异(p〈0.01)。作为生物监测持久性有机污染物的方法,实时定量RT-PCR的灵敏度高于其它已经发表的方法,在污染物的评价上具有很大地应用潜力。This study investigated the effects of Benzo(a)payrene on gene expression of CYP1A1 in liver of Xiphophorus helleri.In this research,vivo method was used to evaluate CYP1A1 gene expression of Xiphophorus helleri in different tissues,and in vitro method was used to determinate CYP1A1gene expression in different concentrations of B(a)P (10-10000ug.L-1)and different induced times(6-48h)by quantitative real-time RT-PCR. Results showed that CYP1A1 were expressed in gill, brain, spleen, liver, kidney, the highest expression in the liver. In vitro, the expression of CYP1A1 gene and B(a)P exposure levels showed a good dose-effect relationship, while that low dose group(10ug.L-1 ) increased significantly (p〈0.05);The activity of liver tissues had decreased with time, but in the same period ,the CYP1A1 gene expression of high dose group (10000ug.L-1)have increased extremely significantly (p〈0.01) compared to the amount of low dose induced. This method is more sensitive compared with other in vitro methods for evaluating Persistent Organic Pollutants. Thus ' applying quantitative real-time RT-PCR to vitro tissue in liver of Xiphophoms helleri has potential application in the evaluation of persistent organic pollutants.
