摘  要:试验旨在通过灌服细菌上清、破碎液建立小鼠生长迟缓模型,为开发具有促生长作用的饲料添加剂提供动物模型。培养大肠杆菌K99,离心收集培养上清,取部分细菌悬液进行超声破碎;试验小鼠随机分为4组(对照组、上清组、破碎组、悬液组),对照组灌服0.2 m L生理盐水,另外3组分别灌服等体积细菌培养上清、破碎液及悬液,每日1次,连续灌服15d;观察小鼠精神状态,记录小鼠体重,测定各脏器指数,采用ELISA方法检测各组小鼠血清IL-6和TNF-α的水平。与空白组相比,其余3组小鼠日增重及脾脏指数均显著降低;肺脏指数显著升高;肝脏、胸腺指数及血清IL-6和TNF-α的水平无显著变化。通过灌服大肠杆菌K99培养上清、破碎液及悬液成功建立了小鼠生长迟缓模型,能够为开发具有细菌毒素灭活作用的中药添加剂提供动物模型。The study was aimed to establish a mouse model of growth retardation by taking bacterial supernatant and broken liquid,which can be used to provide an animal model for the exploitation of growth-promoting animal feed additives. Method: Escherichia coli K99 were cultured to collect culture supernatant and to make bacteria broken liquid by using ultrasonication. Balb/c mice were randomly divided into four groups( control group,supernatant group,ultrasonic crushing group,suspension group). Mice in control group were fed with 0. 2 m L normal saline once a day for 15 days,and mice in other three groups were given respectively equal-volume bacterial culture supernatant,bacterial broken liquid and bacterial suspension. The weight of each mouse and the indexes of visceral organs were measured by using electronic scales,and the levels of IL-6 and TNF-αin serum were detected by ELISA kits. Results showed that both daily gain and spleen index of other three groups were lower than that in control group,but the lung indexs in other three group were significantly increased. The liver index,the thymus index and the levels of IL-6 and TNF-αwere not significantly changed. Results indicated that a mouse model of growth retardation was successfully established by taking bacterial supernatant and broken liquid. And the study may provide an animal model for the screening of growth-promoting animal feed additives from chinese herbs with inactivated effect on bacterial toxin.
