摘 要:为了探索十味降糖散对糖尿病肾病大鼠肾脏的保护作用。选用健康雄性SD大鼠,适应性饲喂1周,随机挑选10只作为空白对照组,其余大鼠高糖高脂饲料饲喂4周+腹腔注射链脲佐菌素复制糖尿病肾病模型。造模成功的大鼠随机挑选10只为灌胃组,10只为模型组,灌胃组灌服十味降糖散汤剂。不同时间检测血糖,第10周末收集各组大鼠24 h尿液,检测尿微量白蛋白;眼眶采血检测血肌酐(CREA)、尿素(UREA);记录体重,取大鼠左肾称重,计算肾脏肥大指数。结果与模型组比较,灌胃组体重升高(P<0.05)。灌胃组肾脏肥大指数与模型组比较有降低趋势,差异有统计学意义(P<0.05);与模型组比较,灌胃组尿微量白蛋白值明显减少(P<0.01);灌胃组的CREA和UREA值极显著下降(P<0.01)。结论:十味降糖散能够降低糖尿病肾病大鼠CREA、UREA指标和尿微量白蛋白值;能够改善糖尿病肾病大鼠体重的降低和缓解肾脏肥大指数增加的趋势,对肾脏具有保护的作用。Objective To investigate the renal protective effects of shiweijiangtang pulvis( SP) on diabetic nephropathy( DN) in rat models. Methods We selected healthy male SD rats and provided them with adaptive feeding for 1 week. Ten rats were randomly selected as the normal control group. The rest of the rats were fed with a high-sucrose and high – fat diet for 4 weeks to establish the DN model by using intra-abdominal strptozotocin injection. Then,the DN rats were randomly divided into two groups,a model control group and a SP group. The rats in the SP group were given gastrogavage for 10 weeks. Fasting blood glucose level was measured once every 4 weeks. The 24-h urine samples of the rats were collected to test for urine microalbumin level in the 10 thweek. Creatinine( CREA) and urea nitrogen( UREA) levels were measured at the end of the 10 thweek. Subsequently,the body weights and weights of the left kidney of the rats were determined to calculate their renal hypertrophy index. Results: Compared with the normal control group,S P group was heavier than model control group( P〈0. 05). Unlike the kidney hypertrophy index of the normal control group. The kidney hypertrophy index of the SP group was significantly decreased compared to the control group( P〈0. 05). The urinary microalbumin level of the SP group was lighter than those of model control group( P〈0. 01). Compared with the model control group,the SP group showed notable decreases in CREA and UREA levels( P〈0. 01). Conclusion Shiweijiangtang pulvis reduced the CREA and UREA and protected their renal function. Additionally,it reduced their weights and delayed the process of kidney hypertrophy.
