摘  要:目的探索以鼻中隔来源软骨细胞作为种子细胞,复合纤维蛋白胶,用于修复大鼠膝关节股骨滑车全层软骨缺损。方法取大鼠鼻中隔软骨细胞,与纤维蛋白胶复合后用于修复大鼠膝关节股骨滑车全层软骨缺损。实验按不同的植人物分为3组:鼻软骨细胞复合纤维蛋白胶(NC+Fibrin)组;单纯纤维蛋白胶(Fibrin)组;单纯缺损(Defect)组。术后6、12周处死实验动物取材。检测并记录Catwalk步态分析、大体观、大体观评分、病理学染色、病理学评分等,评估软骨修复效果。结果术后1、3、6、12周Catwalk步态分析结果显示,NC+Fibrin组大鼠较其他两组可在较短时间内得到更佳的肢体功能恢复。大体观显示NC+Fibrin组的软骨修复表面明显优于其他两组。病理学染色结果显示NC+Fibrin组的修复组织主要以透明软骨为主,而Fibrin组和Defect组的修复组织主要以纤维组织为主。NC+Fibrin组的大体观评分与病理学评分均明显高于Fibrin组和Defect组(P=0.018)。结论以鼻软骨作为种子细胞能够成功修复大鼠膝关节股骨滑车全层软骨缺损。这为组织工程软骨的构建提供了新的思路,也为鼻软骨应用于临床修复关节软骨损伤提供了实验基础。Objective To explore the effect of nasal ehondrocytes combined with fibrin glue as tissue engineering cartilage for repairing full thick articular cartilage defect in a rat model. Methods Nasal chondrocytes were combined with fibrin glue for rat cartilage repair. There are three groups : nasal chondrocytes +fibrin (NC + Fibrin) group; Fibrin group; Defect group. At 6 and 12 weeks after surgery, Catwalk, gross observation, histological evaluation were conducted to evaluate the cartilage repair effects. Results 1, 3, 6, 12 weeks after surgery, the Catwalk results revealed that rats in NC + Fibrin group can obtain a better recovery limb ruction in a shorter time. Gross observation and histological evaluation reveals that NC + Fibrin group led to hyaline - like cartilage repair. While the other two groups generally led to fi- brocartilage repair ( P = 0. 018 ). Conclusion These findings indicate that the nasal chondrocytes can re- pair rat full thick articular cartilage successfully.
