摘 要:目的:观察生姜水煎液对COPD寒饮蕴肺证大鼠的干预作用,并初步探讨其作用机制。方法:烟熏结合气管滴注脂多糖(LPS),饮用冰水加寒冷刺激的方法建立COPD寒饮蕴肺证病证结合大鼠模型,使用生姜水煎液进行干预,通过观察大鼠一般形态,HE染色观察肺组织病理学改变,小动物呼吸机Anires2005检测肺功能,ELISA法检测血液中炎性因子IL-13、IL-10、IL-8、TNF-α以及肺组织中水通道蛋白AQP1、黏蛋白Muc5ac的表达,初步探讨生姜水煎液对此病证的作用机制。结果:与模型组比较,生姜水煎液各给药组均可改善大鼠的一般形态和呼吸功能。并可显著降低大鼠血清中IL-13、IL-10、IL-8、TNF-α的表达;改善肺组织的病理形态,并显著增加肺组织中AQP1的表达、降低Muc5ac的表达。以生姜水煎液高剂量组效果最佳。结论:生姜水煎液可改善COPD寒饮蕴肺证大鼠的一般形态和呼吸功能,其作用机制与降低炎性反应、增加肺组织AQP1蛋白的表达和降低Muc5ac蛋白的表达有关。Objective: To investigate the intervention effect of ginger decoction on chronic obstructive pulmonary disease( COPD)rats with cold fluid in the lung syndrome,and preliminarily explore its mechanism. Methods: COPD rats with cold fluid in the lung syndrome model was established by passive smoking combined with intratracheal instillation of lipopolysaccharide( LPS),and ice drink with cold temperature. The ginger decoction was used to intervene. The general condition of rats was observed. HE staining was used to detect the pathological changes of lung function. Small Animal Ventilator Anires 2005 was used to detect lung function.ELISA was used to detect blood inflammation factor IL-13,IL-10 and IL-8,TNF-α and lung tissue water channel AQP1 protein,sticky MUC5AC protein content. Results: Compared with model group,the administration of ginger decoction group could improve the general condition and respiratory function of rats,and could significantly reduce the expression of IL-13,IL-10 and IL-8,TNF-α in rat's serum; improve the pathology of lung,and significantly increase AQP1 expression,and reduce Muc5ac expression in the lung tissue. The high doses of ginger decoction showed the best effect. Conclusion: Ginger decoction can improve the general condition and ling function of COPD rats with cold fluid in the lung syndrome,whose mechanism is related with reducing inflammation reaction,increasing AQP1 protein and reducing Muc5ac expression in the lung tissue.
