摘  要:目的:研究炮天雄对腺嘌呤制备肾阳虚模型大鼠的治疗作用。方法:采用灌胃腺嘌呤制备大鼠肾阳虚模型,给予不同剂量炮天雄后观察大鼠的一般状态、体重、肛温、饮水饮食量、脏器指数,测定血清中睾酮(T)、环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)、促肾上腺素皮质激素(ACTH)、促甲状腺激素(TSH)、肌酐(Cr)、尿素氮(BUN)、免疫球蛋白G(IgG)等指标水平。结果:与正常组比较,模型组大鼠体重、肛温、饮食量均降低,饮水量增加;大鼠血清中cGMP、ACTH、TSH、BUN、Cr含量均升高,T、IgG含量和cAMP/cGMP比值均降低;肾、甲状腺、肾上腺等脏器指数升高,睾丸、附睾、胸腺、脾等脏器指数降低,差异具有统计学意义(P<0.01或P<0.05),说明腺嘌呤致肾阳虚大鼠模型制备成功。与模型组比较,各给药组大鼠体重和肛温增加,T、IgG含量和cAMP/cGMP比值升高,cGMP、ACTH、TSH、BUN、Cr含量均降低,而cAMP含量没有明显变化;肾、甲状腺、肾上腺等脏器指数均降低,睾丸、附睾、胸腺、脾等脏器指数均升高,差异具有统计学意义(P<0.01或P<0.05)。结论:炮天雄低、中、高剂量组用药对腺嘌呤致肾阳虚模型大鼠具有明显的治疗作用,其作用机制可能与改善下丘脑-垂体-性腺轴(HPG)轴、下丘脑-垂体-甲状腺(HPT)轴和下丘脑-垂体-肾上腺(HPA)轴功能以及免疫功能有关。Objective:To study the therapeutic effect of Paotianxiong(PTX)on adenine induced deficiency of kidney-Yang model rats.Methods:The model rats were given adenine by intragastric administration to establish kidney-YANG deficiency syndrome.During the experiment,the general state,body weight,anal temperature,water and food consumption,also organic index of the rats were recorded.After taking the blood of aorta abdominalis,the levels of testosterone(T),cyclic adenosine monophosphate(cAMP),guanosine monophosphate(cGMP),adrenergic corticosteroids(ACTH),thyrotropin(TSH),creatinine(Cr),urea nitrogen(BUN),immunoglobulin G(IgG)and other indicators were measured in blood serum.Results:Compared to the normal group,the model group appeared the decrease of weight,anal temperature and food consumption,and increase of water consumption.In the meantime,the levels of cGMP,ACTH,TSH,BUN and Cr in blood serum significantly increase(P〈0.01 or P〈0.05),while T,IgG and cAMP/cGMP appeared significantly lower.Among organ index,the index of kidney,thyroid gland and adrenal gland increased,however,testis,epididymis,thymus and spleen decreased,and the differences were statistically significant(P〈0.01 or P〈0.05).It showed that adenine induced kidney-Yang deficiency rat model was satisfactorily replicated.Compared with model group,PTX decoction can increase weight and temperature,and significantly improve T,IgG,cAMP/cGMP,but significantly decrease cGMP,ACTH,TSH,BUN,Cr.A marked decrease in the index of kidney,thyroid gland and adrenal gland,increase in testis,epididymis,thymus and spleen.Conclusion:All the three dosage of PTX decoction can obviously treat the rats with kidney yang deficiency which was induced by adenine.The results also suggested that its mechanism may be related to improve the function of hypothalamic-pituitary-target gland axis(HPG,HPT,HPA)and the immune system.
