摘  要:目的优化小花清风藤药材中总黄酮的提取工艺,并采用比色法测定20批小花清风藤药材中总黄酮的含量。方法在单因素试验基础上,采用响应曲面法考察乙醇浓度、乙醇用量和提取时间对总黄酮提取率的影响,并对其提取工艺进行优化;采用比色法,以芦丁为对照品,于397nm处测定吸光度,计算药材中总黄酮的含量(以芦丁计算)。结果小花清风藤药材中总黄酮的最佳提取工艺为:乙醇浓度79%,乙醇用量56mL,提取时间96min;不同产地小花清风藤药材中总黄酮的含量为3.37%~5.99%。结论所优化的总黄酮提取工艺合理、预测能力强、适用范围广,建立的总黄酮含量测定方法重复性好、结果准确可靠,可为小花清风藤药材的品质评价及相关研究提供参考。OBJECTIVE To optimize the extraction process of the total flavonoids from Sabia parviflora, and to detect the total flavonoids of 20 hatches of S. parviflora by Colorimetrie method. METHODS Baaed on one - way anova, three factors including alcohol concentration,alcohol volume and extracting time which influenced the total flavonoids' content were examined by Response surface methodology. The coloHmetric method was established with rutin as the reference substance. The detection wavelength was 397 nm. The content of the total flavonoids was determined according to rutin. RESULTS The best extraction proeess of the total flavonoids from S. parviflora was as follow:Alcohol concentration was 79%, with the aleohol volume of 56 mL and extracting time of 96 min. The total content of total flavonoids from the different places was between 3.37% - 5.99%. CONCLUSION The extraction process is reasonable and predictable. Its application scope is also wide. This methods is accurate, reliable and has good reproducibility, and can be a reference for the quality evaluation and related research of S. parviflora.
