摘 要:目的研究盐酸戊乙奎醚(PHC)对内毒素诱导大鼠急性肺损伤(ALI)中肿瘤坏死因子-α(TNF-α)及氧化应激的影响。方法按照体重将SD雄性大鼠随机分为4组(每组均10只):正常组(0.9%Na Cl)、模型组(0.9%Na Cl)和低、高2个剂量实验组(PHC:0.3,1.0 mg·kg^(-1))。给药30 min后,尾静脉注射5.0 mg·kg^(-1)脂多糖建立ALI模型。模型制备完成6 h后,处死大鼠,获得肺组织。通过酶联免疫吸附实验测定TNF-α水平;以分光光度计测定肺部组织中过氧化物酶等。结果给药后,正常组、模型组和低、高2个剂量实验组的血清TNF-α分别为(49±10),(106±18),(88±12),(64±7)pg·m L^(-1),血清MDA分别为(2.2±0.6),(3.9±0.3),(3.4±0.6),(2.9±0.3)nmo L·m L^(-1),血清SOD分别为(427±33),(335±43),(356±42),(374±41)U·m L^(-1),模型组与正常组比较,差异均有统计学意义(均P<0.05);低、高2个剂量实验组与模型组比较,差异均有统计学意义(均P<0.05);低剂量组与高剂量组比较,差异均有统计学意义(均P<0.05)。结论盐酸戊乙奎醚预先给药,可降低急性肺损伤大鼠TNF-α含量,减轻炎症反应,提高大鼠氧化应激能力,且高剂量实验组比低剂量实验组效果好。Objective To study the effect of paclitaxyl hydrochloride( PHC) on tumor necrosis factor-α( TNF-α) and oxidative stress in endotoxin-induced acute lung injury( ALI) rats. Methods Male rats were randomly divided into 4 groups( 10 in each group) : normal group( 0. 9% Na Cl),model group( 0. 9% Na Cl),low and high doses experimental groups( PHC: 0. 3,1. 0 mg·kg^(-1)). After administration for 30 min,the tail vein was injected with 5. 0 mg·kg^(-1) lipopolysaccharide to establish the ALI model. After 6 h of preparation,the rats were sacrificed and lung tissue was obtained. TNF-α level was detected by enzyme-linked immunosorbent assay( ELISA),and the expression of peroxidase in lung tissue was measured by spectrophotometer. Results The levels of serum TNF-α in the normal group,model group and low and high dose experimental groups were( 49 ± 10),( 106 ± 18),( 88 ± 12),( 64 ± 7) pg ·m L^(-1),respectively. The levels of serum MDA in the 4 groups were( 2. 2 ± 0. 6),( 3. 9 ± 0. 3),( 3. 4 ± 0. 6),( 2. 9 ± 0. 3)nmo L·m L^(-1),respectively. The levels of serum SOD in the 4 groups were( 427 ± 33),( 335 ± 43),( 356 ± 42),( 374 ± 41) U·m L^(-1). Comparison between model group and normal group,the differences of the factors were significant( all P〈0. 05). Comparison between two doses experimental groups and model group,the differences of the factors were significant( all P〈0. 05). There was significant difference between the low dose group and the high dose group( P〈0. 05). Conclusion Pretreatment with penehyclidine hydrochloride can reduce the content of TNF-α in rats with ALI,reduce the inflammatory reaction and improve the oxidative stress in rats,and the effect of high dose experimental group is better than that of low dose experimental group.
