摘 要:以21个羊肚菌菌株为试材,采用核糖体DNA转录间隔区(ITS)序列分析技术、拮抗试验、菌丝生长特性比较及酯酶同工酶分析的方法,研究了梯棱羊肚菌栽培菌株的遗传特性及亲缘关系。结果表明:梯棱羊肚菌菌丝生长速度为6.90~10.57mm·d^(-1),菌株FY2的生长速度显著高于其它菌株(P<0.05);10个梯棱羊肚菌菌株相异系数在0.53时分为3类,其中FY4菌株与其它菌株之间亲缘关系较远,遗传差异较大,FY1、FY5、FY6、FY7、FY9之间相异系数为0,说明这一类菌株间遗传差异小或为同一菌株;聚类分析结果与拮抗试验结果基本一致。该研究为梯棱羊肚菌遗传信息数据库的建立奠定基础,为优良品种选育和亲缘关系的研究提供依据。Genetic diversity of cultivated strains of Morchella importuna were analyzed by using rDNAITS technique,antagonistic reaction,mycelial growth characteristics and esterase isozyme analysis.The results showed that the mycelial growth rate of M.importuna was 6.90-10.57 mm·d^-1,and strain FY2 was higher than other tested strains(P〈0.05);cluster analysis demonstrated that the 10 M.importuna were divided into 3 groups at the dissimilarity coefficient of 0.53.FY4 had the farthest genetic relationship away from other strains.The similarity of esterase isozyme band of FY1,FY5,FY6,FY7,FY9 indicated that they could be considered as the same strain from different isolation.The results of antagonism were similar to esterase isozyme analysis.The aim of this study was to lay a foundation for the establishment of genetic database,which could provide evidence for selection and breeding of eminent cultivars and genetic relationship in M.importuna.
