摘 要:生长素是植物中非常重要的调控激素,ABP1在很多生长素的调控作用中都起到关键作用,研究证明生长素结合蛋白与信号转导有关,但其具体转导路径还不清楚,本研究以已知拟南芥生长素结合蛋白(ABP1)为模板,在禾本科二穗短柄草、大麦、水稻、高粱、谷子和玉米6个物种中鉴定出10个ABP1蛋白。通过序列保守性分析,我们了解到这些蛋白在C端均有一个保守结构域和典型的保守基序Motif2、Motif3,并且对其进行选择压力分析只得到1个氨基酸位点受到显著正选择。有趣的是水稻Os12g34460基因十分特殊,它在进化树中发生显著分歧且丢失了C端的保守结构域,在亚细胞定位中也是有别于其它所有蛋白。最后,我们以玉米为代表进行蛋白互作分析,发现ABP1与ABP4和ABP5存在密切的关系。本研究使以ABP1为代表的生长素结合蛋白相关性质更加具体清晰,为揭示其作为重要信号的具体传导机制提供参考。Auxin is a very important regulation hormone in plants, and ABP1 plays a key role in the regulation of many auxin. Studies have proved the correlation between auxin binding protein and signal transduction, but the specific transduction way has been still unclear. Using the known Arabidopsis auxin binding protein(ABP1) as a template, the study identified 10 ABP1 proteins in six species of gramineae, including gramineous Brachypodium distachyon, barley, rice, sorghum, millet and maize. Through sequence conservative analysis, we understood that all these proteins had a conserved domain at the C-terminus and a typical conserved motif Motif2, Motif3, and only one amino acid site was significantly selected through the selection pressure analysis. Interestingly, the special rice gene Os12 g34460 was very special. It significantly diverged in the phylogenetic tree, lost the C-terminal conserved domain, and showed difference from all other proteins in subcellular localization. Finally, protein interaction was analyzed on maize and the result indicated close relationships between ABP1 and ABP4 as well as ABP5. Through this study, the correlation properties of auxin binding proteins represented by ABP1 were more specific and clear,which might provide references in reveling it as a specific conduction mechanism of important signals.
