摘 要:本研究以2001年至2015年间审定的16个向日葵主要品种为材料,利用22对SSR荧光标记结合毛细管电泳检测法进行了DNA指纹图谱的构建和遗传关系分析。22对SSR引物共检测出48个多态性片段,大小在100~341 bp之间。每个SSR位点变异范围为2~4个等位基因,平均检测到多态性位点2.18个。多态性信息指数的变化幅度为0.706~0.924,平均值为0.887。对48个等位基因片段大小进行数据记录,建立了各品种特定的SSR指纹图谱。其中的一个引物组合ORS338/ORS229/ORS230/ORS959/ORS316可区分所有的供试材料,为品种鉴别提供依据。聚类分析中,16个向日葵品种的遗传相似系数在0.500 0~0.958 3之间,平均为0.668 1,表明这16个向日葵品种的遗传多样性不够丰富。此工作的开展满足了目前日益增长的品种鉴定的需要,为今后采用SSR荧光标记检测技术进行向日葵品种资源的指纹图谱构建及遗传关系分析提供了依据。Using sixteen sunflower cultivars as materials examined between 2001 and 2015, the study carried out the construction and genetic relationship analysis of DNA fingerprint based on 22 pairs of SSR fluorescent markers combined with capillary electrophoresis test. A total of 48 polymorphic fragments were detected in 22 pairs of SSR primers, and the size was between 100 bp to 341 bp. The variation range of each SSR site was 2 to 4 alleles, with an average detection of 2.18 polymorphism sites. The range of the polymorphism information index was 0.706 to 0.924 with an average of 0.887. The size of 48 alleles were recorded to establish the specific SSR fingerprints of each variety. One of the primer combinations ORS338/ORS229/ORS230/ORS959/ORS316 could distinguish all the test materials which provided an important basis for variety identification. The genetic similarity coefficients of 16 sunflower cultivars ranged from 0.500 0-0.958 3 with an average of 0.668 1, which indicated that there was no abundant genetic diversity among the 16 sunflower cultivars. The work meets the increasing demand of variety identification now, and might provide evidence for the construction of fingerprint map and the genetic relationship analysis of sunflower cultivars using SSR fluorescent marker detection technology in the future.
