摘 要:目的探讨长链非编码RNA(lncRNA)尿路上皮癌胚抗原1(UCA1)在食管鳞癌患者血清中的表达及潜在的临床意义。方法收集96例食管鳞癌患者血清样本,同时选取96例年龄、性别匹配的健康体检者作为健康对照组,其中12例食管鳞癌患者行术前、术后比较。采用实时荧光定量PCR的方法检测lncRNA UCA1在食管鳞癌血清中的表达,并对食管鳞癌患者术前、术后进行比较。分析食管鳞癌患者血清lncRNA UCA1与其临床病理参数的关系,采用受试者工作曲线(ROC)对血清lncRNA UCA1的诊断效能进行评价。结果与健康对照组比较,食管鳞癌患者血清中lncRNA UCA1的表达升高,且患者术后较术前表达下降,差异有统计学意义(P<0.001)。血清lncRNA UCA1诊断食管鳞癌的ROC曲线下面积(AUC)为0.816,灵敏度和特异度分别为77.1%和76.0%。此外,与传统肿瘤标志物鳞状细胞癌相关抗原(AUC=0.679)比较,lncRNA UCA1对食管鳞癌诊断效能更高(P<0.001)。结论血清lncRNA UCA1可作为食管鳞癌早期诊断及术后监测潜在的新型肿瘤标志物。Objective To explore the expression of serunm long non-coding RNA(lnc-RNA) urothelialcarcinoma associated 1(UCA1) in esophageal squamous cell carcinoma(ESCC) and its clinical significance. Methods A total of 96 ESCC cases and another 96 age-and gender-matched healthy controls were involved. The serum samples were collected to determine the expression of lncRNA UCA1 with qRT-PCR. The lncRNA UCA1 expression of 12 ESCC patients were compared before and after operation. The correlation between UCA1 expression and clinical pathological parameters of ESCC were analyzed,and the diagnostic value of serum lncRNA UCA1 was evaluated with the receiver operating characteristic curve(ROC). Results Compared with controls, ESCC patients had upregulated lncRNA UCA1 expression,which decreased after operation(P〈0.001). In addition, the area under the curve(AUC) was 0.816, providing a sensitivity of 77.1% and a specificity of 76.0%. Moreover, lncRNA UCA1 was statistically superior to squamous cell carcinoma antigen(AUC =0.679) in discriminating ESCC patients from healthy controls(P〈0.001). Conclusion Serum lncRNA UCA1 may serve as a potential tumor biomarker for the early diagnosis and postoperative monitoring of ESCC.
