摘 要:【目的】观察姜黄素预处理对肺缺血再灌注损伤模型大鼠炎性细胞因子肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6),抗炎性细胞因子白细胞介素10(IL-10)水平及髓过氧化物酶(MPO)活性的影响,探讨其作用机制。【方法】将48只健康成年雄性SD大鼠随机分为假手术组。模型组,姜黄素低、高剂量组等4组,每组12只。假手术组大鼠于麻醉前2 h腹腔注射等体积生理盐水,开胸后左肺门不进行夹闭,旷置240 min;模型组大鼠于麻醉前2 h腹腔注射等体积生理盐水,左肺门夹闭,左肺不张60 min再灌注180 min;姜黄素低、高剂量组分别于麻醉前2 h腹腔注射姜黄素50、200 mg/kg,行夹闭左肺门6 min再灌注18 min。造模结束后,采用比色法检测肺组织MPO活性,采用酶联免疫吸附(ELISA)法测定肺组织匀浆液中细胞因子TNF-α、IL-6、IL-10的含量。【结果】与假手术组比较,模型组大鼠MPO活性及TNF-α、IL-6含量升高,IL-10含量降低(均P<0.05)。与模型组比较,姜黄素低、高剂量组大鼠MPO活性及TNF-α、IL-6含量降低,IL-10含量升高(P<0.05),其中姜黄素高剂量组效果更为明显(P<0.05)。【结论】姜黄素预处理可能通过下调炎性细胞因子TNF-α、IL-6水平,上调抗炎性细胞因子IL-10,抑制MPO活性,从而减轻大鼠肺缺血再灌注损伤。Objective To observe the effects of curcumin pretreatment on the levels of inflammatory cytokinestumor necrosis factor α(TNF-α),interleukin(IL)-6 and anti-inflammatory cytokine IL-10,and myeloperoxidase(MPO)activity in rats with lung ischemia-reperfusion injury, and to investigate its therapeutic mechanism.Methods Forty-eight healthy adult male SD rats were randomly divided into 4 groups,namely sham-operationgroup,model group,and low-and high-dose curcumin groups,12 rats in each group. The rats in the sham-operation group were given intraperitoneal injection of the same volume of normal saline 2 h before anesthesia,andthen were given thoracotomy but no occlusion of the left hilus pulmonis for 240 min. The rats in the model groupwere given intraperitoneal injection of the same volume of normal saline 2 h before anesthesia,and then were giventhoracotomy and occlusion of the left hilus pulmonis,resulting into left pulmonary atelectasis for 60 min followingby reperfusion for 180 min. The rats in low-and high-dose curcumin groups were intraperitoneal injection of 50,200 mg/kg of normal saline 2 h before anesthesia,and then were given the same performance as that of the modelgroup. At the end of the experiment, MPO activity in rat lung tissues was detected with colorimetry, and thecontents of TNF-α, IL-6 and IL-10 in homogenate of rat lung tissues were determined by enzyme-linkedimmunosorbent assay(ELISA). Results Compared with the sham-operation group,MPO activity and the contentsof IL-6,TNF-α were significantly increased while IL-10 content was decreased in the model group(P〈0.05).Compared with the model group,MPO activity and the contents of IL-6,TNF-α were significantly decreasedwhile IL-10 content was increased in low-and high-dose curcumin groups(P0.05),and the efficacy of thehigh-dose curcumin group was stronger(P0.05).
