摘 要:【目的】观察自然铜与硫氢化钠(NaHS)对骨髓间充质干细胞向成骨细胞分化的影响,初步探讨自然铜发挥"接骨疗伤"功效的可能有效成分。【方法】培养大鼠骨髓间充质干细胞并诱导其转化为成骨细胞,以自然铜与NaHS分别进行干预,采用钙钴法观察成骨细胞碱性磷酸酶(ALP)表达,采用茜素红染色法观察成骨细胞钙结节表达,并对阳性表达的ALP和钙结节进行积分光密度(IOD)的测定。【结果】ALP染色结果显示:与对照组比较,NaHS组ALP表达水平于7、14 d显著增加,自然铜组ALP表达水平于3、7、14 d显著增加(P<0.05);与NaHS组比较,自然铜组3、14 d ALP表达水平显著增加(P<0.05)。茜素红染色结果显示:与对照组和NaHS组比较,自然铜组14 d时钙结节表达水平显著增加(P<0.05)。【结论】自然铜可促进骨髓间充质干细胞向成骨细胞分化,且二硫化亚铁可能是自然铜起"接骨疗伤"作用的有效成分。Objective To observe the effects of Pyritum and Sodium Hydrosulfide(NaHS)on the differentiation of bone marrow mesenchymal stem cells into osteoblasts,and to explore the effective component for reuniting bone and healing sore. Methods The bone marrow mesenchymal stem cells were cultured and induced into osteoblasts,and then were intervened by Pyritum and Na SH respectively. Alkaline phosphatase(ALP)and calcium nodules were observed by Gomori staining method and alizarin red staining method,respectively,and then the integrated optical density(IOD)values of positive results of ALP and calcium nodules were determined. Results ALP staining results showed that the expression level of ALP in NaHS group was increased on culturing day 7,14, and ALP expression level was increased in Pyritum group on culturing day 3,7,14,the difference being significant as compared with that in the blank control group(P 〈 0.05). Compared with NaHS group,ALP expression level was increased in Pyritum group on culturing day 3, 14(P 〈 0.05). Alizarin red staining results showed that the expression level of calcium nodules in Pyritum group on culturing day 14 was increased as compared with that in the blank control group and NaHS group(P 〈 0.05). Conclusion Pyritum can promote the differentiation of bone marrow mesenchymal stem cells into osteoblasts,and ferrous disulfide(FeS_2)may be an effective component of Pyritum for reuniting bone and healing sore.
