摘　　要：目的探讨低密度脂蛋白受体相关蛋白-1(low-density lipoprotein receptor-related protein-1,LRP1)与黑素瘤转移的关系。方法沉默黑素瘤细胞A375的LRP1基因,测试划痕实验和Transwell实验;利用免疫组织化学(IHC)检测组织芯片上正常皮肤组织和黑素瘤皮肤组织的LRP1蛋白表达情况。结果黑素瘤A375细胞中沉默LRP1基因后,黑素瘤A375细胞的迁移能力和转移能力减弱;组织芯片免疫组织化学结果发现LRP1蛋白在黑素瘤皮肤组织中相对于正常皮肤组织表达增高。结论 LRP1蛋白可以促进黑素瘤A375细胞的转移;LRP1蛋白可能成为黑素瘤潜在的肿瘤标记物以及黑素瘤诊断治疗的靶蛋白。Objective To study the effect of LRP1 protein on migration of melanoma. Methods Migration and metastasis of A375 melanoma cells were evaluated after silencing the LRP1 gene. Immunohistochemistry （IHC） was used to evaluate expression levels of LRP1 protein in cutaneous malignant melanoma and normal skin. Results The si- lenced LRP1 gene decreased migration and metastasis of A375 cell. Results of mmu- nohistochemistry （IHC） showed that malignant melanoma skin expressed higher levels of LRP1 protein in comparison with normal control. Conclusion LRPI can stimulate migration and metastasis of A375 melanoma cells. LRP1 protein may be a potential marker of melanoma and the target protein in the diagnosis and treatment of the mela- noma.
