摘 要:目的探究中药葛花提取物甲胺鸢尾素对大鼠心肌缺血损伤后相关炎性因子的影响机制。方法将大鼠分为4组,实验1、2组采用不同剂量甲胺鸢尾素灌胃,模型组及空白组等量生理盐水灌胃,10d各组(空白组除外)均皮下多点注射盐酸异丙肾上腺素0.4mg/100g,制备异丙肾上腺素致大鼠心肌损伤模型。造模后,取各组大鼠相应部位材质,检测并对比分析各组大鼠的TNF-α、IL-1β、Bcl-2及NF-κB蛋白的变化情况。结果与空白组对比,模型组大鼠及实验组血清TNF-α、IL-1β、Bcl-2、NF-κB明显升高(P<0.05);与模型组对比,实验1、2组血清TNF-α、IL-1β、NF-κB明显降低(P<0.05),Bcl-2明显升高(P<0.05),其中实验1组比实验2组降低/升高更加明显(P<0.01)。结论甲胺鸢尾素对异丙肾上腺素致大鼠缺血再灌注损伤能起到保护作用,能够有效抑制TNF-α、IL-1β炎症因子的释放,上调Bcl-2,抑制NF-κB蛋白表达。Objective To explore the traditional Chinese medicine Pueraria flower extract methylamine Irisolidone on rat myocardial ischemia injury related inflammatory factors influence mechanism.Methods The rats were divided into 4 groups,experimental group 1,2 with different doses of methylamine Irisolidone gavage,model group and blank control group normal saline,10 dgroup(except the blank group)were subcutaneous injection of isoprenaline hydrochloride 0.4 mg/100 g,preparation of isoproterenol induced myocardial injury model in rats.After modeling,the rats take corresponding parts of materials,testing and comparative analysis of the rats of TNF-α,IL-1β,the changes of Bcl-2 and NF-κB protein.Results compared with the blank group,model group rats serum TNF-α,IL-1 beta,Bcl-2,NF-κB increased significantly(P〈0.05),while the experimental group serum TNF-alpha,IL-1 beta,Bcl-2,NF-κB decreased significantly(P〈0.05).Compared with the model group,the group 1,2 serum TNF-α,IL-1β,Bcl-2,NF-κB significantly decreased(P〈0.05),the experimental group 1 than group 2 significantly decreased(P〈0.01).Conclusion The effects of methamine tectoridin on ischemia-reperfusion injury in rats induced by isoprenaline play aprotective role.It can effectively inhibit the release of TNF-αand IL-1βinflammatory factors,and up-regulate Bcl-2 and inhibit the expression of NF-κB protein.
