摘  要:目的:探讨半胱氨酸丰富跨膜成骨蛋白调控因子1(Crim1)在肥大心肌组织中的表达情况以及血管紧张素受体1型(AT1R)对其蛋白表达的调控作用。方法:将220~250g SD清洁级雄鼠随机分为4组:腹部假手术+等体积0.9%氯化钠溶液灌胃(对照组),腹主动脉缩窄术+等体积0.9%氯化钠溶液灌胃(AAC组),腹主动脉缩窄术+替米沙坦灌胃(3.57mg·kg^(-1)·d^(-1))(AAC+T组),腹部假手术+替米沙坦灌胃(3.57mg·kg^(-1)·d^(-1))(T组)。超声心动图测量各组大鼠左心室各室壁厚度、左心室内径,并计算左心室质量(校正值);HE染色检测各组大鼠心室肌细胞横截面积,Western blot、Crim组织化学检测各组大鼠心室肌细胞Crim1蛋白的表达。结果:腹主动脉缩窄术诱导的大鼠肥大心室肌中的Crim1蛋白表达下调,而替米沙坦可抑制此下调过程。结论:肥大心肌组织中Crim1蛋白表达下调,AT1R信号通路可能介导了肥大心肌组织中Crim1蛋白表达的调控。Objective:To investigate the expression of cysteine-rich motor neuron 1(Crim1)protein in hypertrophic myocardial tissues and the effect of angiotensin receptor 1 type(AT1R)on Crim1 protein expression.Method:220-250 g Sprague-Dawley(SD)rats were randomly divided into 4 groups:control group(sham operation),AAC group(abdominal aortic constriction),AAC+T group(abdominal aorta constriction plus Telmisartan,3.57 mg·kg^-1·d^-1),Telmisartan group(sham operation plus Telmisartan,3.57 mg·kg^-1·d^-1).The left ventricular(LV)wall thickness,LV internal dimension and corrected LV mass were measured by echocardiography.The cross-sectional area of cardiomyocytes was monitored by HE staining.Crim1 protein expression was detected by Western blotting and immunohistochemical staining.Result:The expression of Crim1 protein was significantly reduced in AAC-induced hypertrophic myocardial tissues in SD rats,while Telmisartan prevented this effect.Conclusion:The expression of Crim1 protein is down-regulated in hypertrophic myocardial tissues,which might be regulated by AT1R.
