摘  要:目的探讨橙皮素干预对百草枯(PQ)致大鼠肺损伤的拮抗作用及其对氧化指标的影响。方法 140只Wistar雄性大鼠随机分为正常对照组、橙皮素对照组、PQ模型组、吡非尼酮对照组及橙皮素低、中、高剂量治疗组,每组20只。正常对照组和橙皮素对照组不染毒,其他各组均以50 mg/kg PQ一次性灌胃染毒,24 h后对各组进行药物干预,每天灌胃给药1次。分别于染毒后第7、28天每组随机处死10只大鼠,观察并比较各组大鼠肺组织病理学改变,检测每组肺组织中丙二醛(MDA)水平及超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)和总抗氧化能力(T-AOC)活力。结果组织病理学检查发现,吡非尼酮对照组及橙皮素各治疗组大鼠肺损伤情况减轻。正常对照组与橙皮素对照组大鼠肺组织中各氧化指标水平无明显差异(P>0.05);与正常对照组比较,PQ染毒后肺组织中MDA水平升高,SOD、CAT和GSH-Px活力降低;橙皮素干预后MDA水平降低,SOD、CAT和GSH-Px活力出现不同程度升高,差异有统计学意义(P<0.05或P<0.01)。结论橙皮素可能通过抑制氧化损伤拮抗PQ中毒大鼠肺损伤。Objective To understand the protective effect of hesperetin on oxidative damage in rats lung injury induced by paraquat. Methods A total of 140 male Wistar rats were randomly divided into normal control group, hesperetin control group,paraquat model group, pirfenidone control group, low, medium and high dose of hesperetin treatment group, 20 rats in each group. All groups received a single administration of paraquat at 50 mg/kg body weight through gavage except for the normal control group and the hesperetin control group, 24 hours after the treatments, the rats in each group were given drug intervention once daily.A total of 10 rats were randomly selected respectively at the 7 th day and the 28 th day after exposure to paraquat. The pathological examination of the lung tissue in each group was conducted. The level of malondialdehyde(MDA)and the activities of superoxide dismutase(SOD), catalase(CAT), glutathione peroxidase(GSH-Px) and total antioxidant capacity(T-AOC) were determined in all groups. Results Histopathological examination indicated that pirfenidone and hesperetin treatment groups could reduce the lung injury; There was no significant difference between normal control group and hesperetin control group in the levels of the oxidation indexes in lung tissues(P 0.05). Compared with the normal control group, the level of MDA in lung tissue significantly increased in paraquat exposure groups(P〈0.01) and the activities of SOD,CAT and GSH-Px significantly decreased(P 0.05). After the intervention of hesperetin, the level of MDA significantly decreased(P〈0.05, P〈0.01) and the activities of SOD, CAT and GSH-Px significantly increased in varying degrees(P〈0.05, P〈0.01). Conclusion Hesperetin has protective effect on the lung tissue injury induced by paraquat in rats by inhibiting the oxidative damage and enhancing the antioxidant capacity.
