摘  要:目的探讨长链非编码(lnc)RNA LOC101927019在大气PM_(2.5)致人支气管上皮细胞(16HBE细胞)炎症反应中的作用。方法将采集的广州市大气PM_(2.5)制备成混悬液染毒16HBE细胞,以荧光定量PCR检测不同浓度(25~100μg/ml)PM_(2.5)染毒细胞炎症因子IL-1β基因及lnc RNA LOC101927019的表达水平。采用RNA干扰技术敲低16HBE细胞中LOC101927019的表达,检测100μg/ml PM_(2.5)染毒si RNALOC101927019-16HBE细胞中IL-1β基因的表达水平。结果与对照组比较,PM_(2.5)染毒16HBE细胞IL-1β及LOC101927019的表达上调(P<0.01);与对照组比较,16HBEsi RNALOC101927019细胞LOC101927019的表达明显下降(P<0.05);与PM_(2.5)染毒转染阴性对照组相比,PM_(2.5)染毒16HBE-si RNALOC101927019细胞IL-1β基因的表达明显下降(P<0.01)。结论 lnc RNALOC101927019在PM_(2.5)暴露染毒的细胞中高表达,敲低lnc RNALOC101927019表达可抑制IL-1β的分泌,lnc RNALOC101927019可能在PM_(2.5)致细胞炎症反应中起促炎作用。Objective To investigate the role of long non-coding RNA LOC101927019 in inflammatory response in the normal human bronchial epithelial cells(16 HBE) exposed to PM2.5. Methods Ambient PM2.5 was sampled in Guangzhou.Mixed PM2.5 suspension was prepared. Expression levels of IL-1β and lnc RNA LOC101927019 gene in 16 HBE cells exposed to PM2.5 with different concentrations(25-100 μg/ml) were detected by q PCR.RNA interference was used to reduce the expression of LOC101927019 in 16 HBE cells, the expression level of IL-1β was detected in si RNALOC101927019-16 HBE exposed to 100μg/ml PM2.5. Results Compared with the control, the expression of IL-1β and lnc RNALOC101927019 increased in 16 HBE cells exposed to PM2.5(P〈0.01). The expression of lnc RNALOC101927019 in si RNALOC101927019-16 HBE cells decreased compared with the non-transfection 16 HBE cells(P〈0.05). The expression of lnc RNALOC101927019 in si RNALOC101927019-16 HBE cells exposed to 100 μg/ml PM2.5 decreased significantly compared with the non-transfection 16 HBE cells exposed to100 μg/ml PM2.5(P〈0.01). Conclusion Lnc RNALOC101927019 is highly expressed in 16 HBE cells exposed to PM2.5, and knock-down of lnc RNALOC101927019 can inhibit the secretion of the inflammatory factor interleukin IL-1β.Lnc RNALOC101927019 may promote inflammation induecd by PM2.5.
