摘 要:应用蛋白质组学技术(i TRAQ)分析仔猪耳廓血液γ-干扰素(IFN-γ)变化过程中的差异表达蛋白,探讨IFN-γ在耳廓微循环中的机制和作用。通过3次生物学重复试验,共鉴定出224种显著差异表达蛋白质,与对照组相比,其中122种蛋白的表达量上调,102种蛋白的表达量下调。生物信息学分析提示这些蛋白主要参与脱氧核糖核酸酶活性调控、血小板激活和聚集等过程。代谢通路分析显示,这些蛋白主要参与核糖体、剪切体和内质网蛋白等多种信号通路,涉及调控细胞骨架、细胞黏附性及细胞因子转录等多种生理活动,初步鉴定核因子κB在IFN-γ的调控中发挥了重要的作用。用生物高通量技术检测了IFN-γ注射后差异表达蛋白质在仔猪耳廓微循环中的表达,为疾病监控和治疗提供新型的生物标志物和靶向药物作用位点奠定了基础。To reveal the mechanisms of interferon-γ(IFN-γ) modulating auricular vascular microcirculation in piglets,isobaric tags for relative and absolute quantitation(i TRAQ) quantitative proteomics technology was used to analyze the differentially expressed proteins followed by serum IFN-γelevation.A total of 224 different proteins were identified in three biological replications,of those proteins,122 were up-regulated,and 102 were down-regulated compared with the control.Bioinformatics analysis showed these proteins were related to the activity of deoxyribonuclease and platelet activation and aggregation.The GO and KEGG pathways analysis also revealed these proteins were involved in signal pathways of ribosome,spliceosome and protein processing in endoplasmic reticulum,which confirmed in physiological functions of cytoskeleton,cell adhesion and cytokine transcription.NF-κB pathway was preliminary identified plays a significant role in response to IFN-γ.A high throughput technique was used to detect expression of the differential proteins in piglets auricular microcirculation after IFN-γ injection,and the results lays the foundation for monitoring and treating diseases to provide a new biomarker and target site for diseases therapy.
