摘 要:为研究N-乙酰半胱氨酸(NAC)在镉致PC12细胞凋亡线粒体途径中的保护作用,本试验用10mol/L醋酸镉和100 mol/L NAC单独或联合处理PC12细胞24 h后,用MTT法检测细胞存活率;用流式细胞术检测细胞凋亡率;Western-blot法检测细胞色素C(cytochrome c,Cyt C)的释放以及Bcl-2、Bax、Cleaved caspase-3和Cleaved PARP蛋白的表达量。结果,NAC能显著或极显著抑制镉引起的细胞存活率下降,凋亡率上升,Cyt C从线粒体释放进入胞浆中,Bcl-2/Bax比值下降,Cleaved caspase-3和Cleaved PARP蛋白的表达量提高(P〈0.05或P〈0.01)。上述结果说明,NAC在镉致PC12细胞凋亡线粒体途径中具有保护作用。To investigate the protective effect of N-acetylcysteine(NAC) on mitochondrial apoptotic pathway induced by cadmium(Cd),PC-12 cells were exposed to Cd(10 mol/L) and/or 100 mol/L NAC for 24 h.Then,cell viability was assayed by MTT assay,cell apoptotic rate was measured by flow cytometry,the release of cytochrome c(Cyt C) and the protein levels of Bcl-2,Bax,cleaved caspase-3 and cleaved PARP were measured by Western-blot.In result,Cd-induced decrease of cell viability, increase of apoptotic rate,release of Cyt C,reduction of the B-cell lymphoma 2(Bcl-2)/Bcl-2 associate X protein(Bax) ratio,as well as increase of Cleavedcapase-3 and Cleaved PARP protein levels were inhibited by NAC significantly(P〈0.05 or P〈0.01).These findings suggest that NAC can provides protection against Cdinduced apoptosis via a mitochondrial apoptotic pathway in PC12 cells.
