摘  要:侧根是根系锚定植株、吸收水分和矿质养分等的重要器官,与作物生产密切相关。本研究通过逆转录PCR(reverse transcription PCR,RT-PCR)技术克隆获得1个甘蓝型油菜(Brassica napus)MYB(v-myb avian myeloblastosis viral oncogene homolog)类转录因子,命名为BnMYB193(Gen Bank登录号:XP_013674792.1),其ORF长度为1 083 bp,编码361个氨基酸;生物信息学分析表明,BnMYB193基因编码蛋白的相对分子质量为40.63 k D,含有2个保守的MYB重复结构域,为典型的植物R2R3-MYB转录因子;qRT-PCR分析表明,BnMYB193基因在油菜苗期的根中优势表达,且在侧根中的表达量明显高于主根,表明该基因可能对侧根发育具有调控作用;激素诱导表达分析表明,该基因的表达明显受生长素(auxin,IAA)、脱落酸(abscisic acid,ABA)、赤霉素(gibberellin,GA3)和细胞分裂素(cytokinins,6-BA)的诱导,暗示该基因可能参与植物激素信号转导过程。酵母(Saccharomyces cerevisiae)单杂交实验结果显示,该转录因子具有转录激活活性。此研究为进一步探究BnMYB193基因的功能和油菜侧根发育的调节机理提供了参考资料。Lateral root is a key organ for plant anchorage, acquisition of water and nutrient uptake etc, which is important for crop productivity. In this study, a MYB (v-myb avian myeloblastosis viral oncogene homolog) transcription factor named BnMYB193 (GenBank accession No. XP_013674792.1) was cloned from Brassica napus by reverse transcription PCR (RT-PCR) method. The ORF full-length ofBnMYB193 gene was 1 083 bp, encoding 361 amino acids. Bioinformatics analysis showed that the molecular weight of its protein was 40.63 kD, which contained 2 conserved MYB repeats, indicating it was a typical R2R3-MYB transcription factor, qRT-PCR analysis showed that BnMYB193 preferentially expressed in roots of B. napus at seedling stage with the relative higher expression level in lateral roots as compared with taproots, indicating it may be involved in regulating lateral root development. In addition, the expression level of BnMYB193 gene was obviously up-regulated by auxin (IAA), abscisic acid (ABA), gibberellin (GA3) and cytokinins (6-BA) inductions, implying a possible role of this gene in plant hormone signal transduction pathways. Yeast (Saccharomyces cerevisiae) one hybrid analysis showed that the candidate protein had transcription activity. These results provide reference for exploring the function of BnMYB193 gene and the regulating mechanism of B. napus lateral root development.
