摘 要:解偶联蛋白(uncoupling protein,UCP)在动物机体能量代谢和脂类代谢方面发挥着重要作用。为探讨UCP2和UCP3基因作为黑安格斯牛生长性状候选基因的可能性,及寻找与黑安格斯牛生长性状相关的分子标记,本研究以宁夏地区739头黑安格斯牛(Bos taurus)为研究对象,利用Sequenom SNP分型技术,对UCP2和UCP3基因进行SNP分型,研究其多态性,并分析其多态位点不同基因型与黑安格斯牛生长性状的关联性。通过遗传多态性分析发现,4个SNPs(UCP2-655、UCP3-5465、UCP3-7921和UCP3-8018)均处于Hardy-Weinberg平衡状态(P>0.05),除UCP2-655位点属于中度多态外(0.25<PIC<0.5),其余3个位点均属于低度多态(PIC<0.25);利用Popgene32软件对4个位点的等位基因频率和基因型频率进行分析发现,UCP2-655位点优势等位基因A的频率为0.809;UCP3-5465位点优势等位基因G的频率为0.907;UCP3-7921位点优势等位基因G的频率为0.978;UCP3-8018位点优势等位基因C的频率为0.978;关联性分析结果表明,在本实验群体中,UCP3-5465位点对黑安格斯牛管围的效应达到显著水平(P<0.05),GG型个体均值在管围方面显著高于AA型和AG型个体均值。UCP3-7921位点和UCP3-8018位点对黑安格斯牛体重的效应均达到显著水平(P<0.05),UCP3-7921位点GG型个体均值在体重方面显著高于GC型个体均值,UCP3-8018位点CC型个体均值在体重方面显著高于CT型个体均值。综上,UCP3基因对黑安格斯牛部分体尺性状有显著影响,可以尝试将UCP3基因作为影响黑安格斯牛生长性状的主效基因,为黑安格斯牛选育工作提供理论依据。The aim of this study was to investigate the possibility of UCP2 and UCP3 as candidate gene of growth traits in Black Angus cattle (Bos taurus) and to find molecular markers related to the growth traits of Black Angus cattle. 739 Black Angus cattle in Ningxia area were chosen as study animals, Sequenom SNP was applied to detect the polymorphisms of UCP2 and UCP3 gene, and analyzed the genotypes of polymorphic loci correlation of growth traits in Black Angus cattle. Four SNPs (UCP2-655, UCP3-5465, UCP3-7921 and UCP3-8018) were found to be in Hardy-Weinberg equilibrium by genetic polymorphism analysis(P〉0.05),except that UCP2-655 was moderate polymorphism (0.25〈PIC〈0.5), and the other three loci were low polymorphisms (PIC〈0.25). Using Popgene 32 software to analyze the allele frequency and genotype frequency of 4 loci, this study found that the frequency of dominant allele A of UCP2-655 locus was 0.809; UCP3-5465 locus dominant allele G frequency was 0.907; UCP3-7921 dominant allele G frequency was 0.978; The prevalence of allele C at UCP3-8018 was 0.978. Association analysis showed that the UCP3-5465 SNP had significant effects on the Black Angus cattle cannon circumference in this experimental population (P 〈0.05), The cattle with genotype GG had greater cannon circumference than those with genotype AA and AG. UCP3-7921 and UCP3-8018 had significant effects on body weight of Black Angus cattle (P〈0.05), At the UCP3-7921 locus, the cattle with genotype GG had greater body weight than those with genotype GC, At the UCP3-8018 locus, the cattle with genotype CC had greater body weight than those with genotype CT. In conclusion, UCP3 gene had potential effects on growth traits of Black Angus cattle, Therefore, variations in the UCP3 gene may be useful as major genes for growth traits in Black Angus cattle which would provide a theoretical basis for breeding of Black Angus cattle.
