摘  要:转移抑制素(kisspeptins,KISS1)被认为是哺乳动物繁殖机制的调控者。KISS1基因编码的蛋白,可以影响促性腺激素释放激素的释放,具有调节动物性腺产生性激素与生殖细胞的能力。本研究以黔北麻羊(Capra hircus)为研究对象,对KISS1基因的多态性及其在繁殖相关组织的表达量进行研究,旨在揭示KISS1基因对黔北麻羊产羔性状的影响。结果表明,在黔北麻羊内含子1处检测到3个SNPs,分别为G277C,G436T和T486A,其中在G277C、T486A位点,黔北麻羊3种基因型个体间的产羔数达到显著差异水平。qRT-PCR结果显示,在检测的黔北麻羊的5个组织中KISS1基因均有表达,其中下丘脑的表达量显著高于其他各组织(P<0.05),但子宫、卵巢、输卵管3个组织间的KISS1基因的表达量差异不显著(P>0.05)。结果表明,KISS1基因中的G277C、T486A位点的突变对黔北麻羊的产羔数有一定的关联,对其进行研究可为黔北麻羊高繁殖力品系的培育提供一定的基础。Kisspeptins (KISS1) is considered to be a regulator of mammalian reproductive mechanisms. The protein encoded by KISSI gene can affect the release of gonadotrophic hormone releasing hormones and regulate the production of sex hormone and reproductive cells of animal glands. In this study, the polymorphism and tissue expression of KISS1 gene in hypothalamus-pituitary-gonadal axis was investigated to elucidate the relationship between KISS1 gene and the litter size of Qianbei Ma goat(Capra hircus). The results showed that three SNPs including G277C, G436T and T486A were detected in the intron 1 of Qianbei Ma goat. There were significant difference between the sites G277C and T486A and the three genotypes of the little size of Qianbei Ma Goat. The results of qRT-PCR showed that the expression of KISS1 gene in hypothalamic was much higher than that in other tissues (P〈0.05), but no significant difference in the expression of KISS1 gene was detected in uterus, ovary and fallopian tube (P〉0.05). These results indicated that G277C and T486A loci in KISS1 gene might be associated with the litter size of Qianbei Ma Goat, which lay the foundation for further cultivating high fecundity lines of Qianbei Ma Goat.
