摘  要:绵羊(Ovis aries)卵巢中血管生成因子的表达与其血管生长、微血管密度及卵泡发育密切相关,且对绵羊的生产性能具有重要影响。本研究采用qRT-PCR技术,检测血管内皮生长因子基因(vascular endothelial growth factor gene,VEGF)、促血管生成素/酪氨酸激酶受体信号通路(angiopoietin/tyrosine kinase receptors2,Ang/Tie-2),包括血管生成素-1(angiopoietin-1,Ang-1)、血管生成素-2(angiopoietin-2,Ang-2)、酪氨酸激酶受体-2(tyrosine kinase receptors,Tie-2)在小尾寒羊、湖羊及藏羊卵巢组织中的表达量,探讨其在不同繁殖力绵羊品种间的表达及规律。结果显示VEGF、Ang-1、Ang-2及Tie-2在小尾寒羊、湖羊及藏羊卵巢中表达差异显著(P<0.05),VEGF、Ang-1及Tie-2的表达量与产羔率呈正相关,Ang-2的表达量与产羔率呈负相关,说明促进血管发生的Ang/Tie-2通路对绵羊的排卵率有影响,同时受其他血管生成因子特别是VEGF的调节。本研究为初步揭示绵羊繁殖力差异的遗传背景提供了基础资料。The expression of angiogenic factors in sheep ovaries is closely related to the vascular growth, microvessel density and follicular development, which plays an important role in the reproductive performance of sheep. Firstly, the total RNA in ovaries of different sheep breeds were extracted and their quality control (QC) was conducted by ultraviolet spectrophotometry and agarose gel electrophoresis, which optical density value (OD) was ranged of 1.8〈OD26o/OD28o〈2.0 and its 28s, 18s, 5s bands were observed clearly non-degradable, then the total RNA were transcribed into cDNA and their concentration and OD value were detected by the ultraviolet spectrophotometer for meeting the availability of the experiment. Secondly, the expression levels of vascular endothelial growth factor gene (VEGF), angiopoietin/tyrosine kinase signaling pathway (angiopoietin/tyrosine kinase receptors2, Ang/Tie-2), including angiopoietin-1 (Ang-1), angiopoietin-2 (Ang-2), tyrosine kinase receptors2 (Tie-2) in ovaries of Small-Tailed Han sheep, Hu sheep and Tibetan sheep (Oura-type) were determined by quantitative real-time PCR (qRT-PCR), and expressional differences of angiogenic genes in different breeds were analyzed. Results showed that the expression levels of VEGF, Ang-1, Ang-2 and Tie-2 genes were significantly different among three breeds (P〈0.05). The expression level of VEGF showed significant difference among Tibetan sheep (single lambs), Hu sheep, Small- Tailed Han sheep (multiple lambs) (P〈0.05), in which the expression level of VEGF in ovary of Small-Tailed Han sheep was 3.20 times and 2.72 times as that of Tibetan sheep and Hu sheep, respectively. Similarly, the expression level of Ang-1 in Small-Tailed Han sheep was significantly higher than that of Hu sheep and Tibetan sheep (P〈0.05), and Small-Tailed Han sheep was 2.46 times as that of Tibetan sheep while Hu sheep was 1.82 times as that of Tibetan sheep.
